分心木抗炎活性部位筛选及其化学成分分析

Screening of Anti-Inflammatory Active Part of Diaphragma juglandis Fructus and Analysis of Its Chemical Constituents

摘要 以脂多糖(LPS)诱导的RAW264.7细胞为炎症模型,采用Griess法检测细胞上清液中一氧化氮(NO)水平,采用酶联免疫吸附法(ELISA)检测细胞上清液中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的水平,评价了分心木不同极性部位的体外抗炎活性;进一步通过超高效液相色谱-线性离子阱-静电场轨道阱高分辨质谱(UPLC-LTQ-Orbi-trapHRMS)技术对分心木抗炎活性最高部位的化学成分进行了鉴定。结果表明,与模型组比较,除水部位外,其他部位在一定剂量下均能显著降低细胞上清液中NO、IL-6、TNF-α的水平(P<0.05或P<0.01),水部位在一定剂量下能够显著降低IL-6的水平(P<0.05);总体来看,乙酸乙酯部位的抗炎活性最高,从中共鉴定出44个化学成分,包括13个黄酮类化合物、11个脂肪酸类化合物、10个酚酸类化合物、5个鞣质类化合物、2个醌类化合物、2个萜类化合物和1个香豆素类化合物,为深入研究分心木抗炎作用的药效物质基础奠定了基础。 Taking lipopolysaccharide(LPS)-induced RAW264.7 cell as an inflammatory model,we detected the level of nitric oxide(NO)in the cell supernatant by Griess method,detected the levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)by enzyme-linked immunosorbent assay(ELISA),and evaluated the in vitro anti-inflammatory activity of different polar parts of Diaphragma juglandis Fructus.Moreover,we identified the chemical constituents of the part of Diaphragma juglandis Fructus with the highest anti-inflammatory activity via UPLC-LTQ-Orbitrap HRMS technique.The results show that,compared with model group,the levels of NO,IL-6,and TNF-αin the cell supernatant significantly decrease in the other parts except the water part at a certain dose(P<0.05 or P<0.01),and the level of IL-6 in the cell supernatant significantly decreases in water part at a certain dose(P<0.05).Overall,ethyl acetate part has the highest anti-inflammatory activity.We identified 44 chemical constituents from ethyl acetate part,including 13 flavonoids,11 fatty acids,10 phenol-ic acids,5 tannins,2 quinones,2 terpenoids,and 1 coumarin.This study lays a foundation for the in-depth study on the anti-inflammatory material basis of Diaphragma juglandis Fructus.
