摘要
目的探讨右美托咪定(DEX)在膝关节骨关节炎(KOA)治疗中的价值及其机制。方法SD大鼠随机分为对照组、模型组、治疗组,每组10只。通过形态学染色方法观察各组大鼠将膝关节软骨组织,采用Western blot检测各组软骨组织细胞凋亡及脂酰肌醇-3-激酶-蛋白激酶B-内质网应激(PI3K-AKT-ERS)通路蛋白表达。结果模型组大鼠膝关节软骨破坏显著,关节腔积液IL-6β、TNF-α水平升高;与模型组比较,治疗组大鼠软骨组织结构完整清晰,关节腔积液IL-6β、TNF-α表达水平降低(P<0.05)。与模型组比较,治疗组细胞凋亡率水平显著降低(P<0.05);Western blot检测表明,与模型组比较,治疗组Bcl-2样蛋白4(Bax)、半胱氨酸天冬氨酸蛋白酶3(Caspase3)、激活转录因子4(ATF4)、G蛋白偶联受体78(GPR78)及内质网应激相关蛋白(CHOP)蛋白水平表达显著下调(P<0.05),PI3K、AKT蛋白水平显著升高(P<0.05)。结论右美托咪定通过调控PI3K-AKT-ERS通路抑制关节炎软骨损伤。
Objective To explore the value and mechanism of dexmedetomidine(DEX)in the treatment of knee osteoarthritis(KOA).Methods SD rats were randomly divided into the control group,the model group,and the dexmedetomidine treatment group,10 rats in each group.The knee cartilage tissues of rats in each group were observed by morphological staining method,and the apoptosis of cartilage tissue cells and the expression of PI3K-AKT-ERS pathway proteins in each group were detected by Western blot.Results The cartilage of the knee joint in the model group was significantly damaged,and the levels of IL-6βand TNF-αin the joint cavity fluid were increased;comparing with the model group,the cartilage tissue structure of rats in the treatment group was clear and intact,and the expression levels of IL-6βand TNF-αin the joint cavity fluid were decreased(P<0.05).Comparing with the model group,the apoptosis rate of cells in the treatment group was significantly ldecreaseed(P<0.05).Comparing with the model group,the protein levels of Bax,Caspase3,ATF,GPR78,and CHOP in the treatment group were significantly down-regulated(P<0.05),and the protein levels of PI3K and AKT were significantly up-regulated(P<0.05).Conclusion Dexmedetomidine could inhibit cartilage damage in arthritis by regulating the PI3K-AKT-ERS pathway.
