摘要
目的:基于“以方(药)测靶”“靶标关联”“由标知效”“临床验效”和“传承创新”组成的中医方药整体作用靶标研究模式,精准阐释名医名方芩栀猪胆方在脑病治疗中的作用,发掘其更多潜在新功效。方法:采用热蛋白质组学技术,对芩栀猪胆方在小鼠脑组织中的整体作用靶点进行高通量筛选;利用IPA数据库对芩栀猪胆方作用靶点和下游目标蛋白进行关联研究;进一步运用Genevestigator平台对靶标关联的疾病进行挖掘和分析。结果:共筛选获得芩栀猪胆方水煎液直接作用靶点20个、间接作用靶点18个;通过IPA数据库和Genevestigator平台联合分析其关联疾病发现,这些靶点分别参与阿尔茨海默病、缺血性脑卒中、抑郁症、血管性痴呆等疾病的治疗,提示芩栀猪胆方具有治疗共济失调、脑胶质瘤的潜在新功效。结论:从靶标角度阐明了芩栀猪胆方水煎液治疗脑病的作用机制,进一步支持其在共济失调、脑胶质瘤等中枢神经系统疾病治疗中具有潜在应用价值。
Objective:Based on the research model for integrative effect of TCM,which consist of modules including"detecting targets of the drug","targeted-regulated molecule association","obtaining the efficacy with the regulated molecule","clinical validation of efficacy",and"inheritance and innovation of TCM",this study gave precise explanation for the function of Qinzhi Zhudan Decoction in brain diseases treatment,and reveals its potential new efficacy.Methods:Thermal proteome profiling(TPP)was employed to perform high-throughput screening of the overall targets of Qinzhi Zhudan Decoction in mouse brain tissue.The ingenuity pathway analysis(IPA)database was used to analyze the associations between the targets of Qinzhi Zhudan Decoction and downstream target proteins.Furthermore,the Genevestigator platform was utilized to explore diseases associated with these targets.Results:A total of 20 direct targets and 18 indirect targets of Qinzhi Zhudan Decoction were identified.Joint analysis using the IPA database and Genevestigator platform revealed that these targets were involved in the treatment of Alzheimer′s disease,ischemic stroke,depression,vascular dementia,and other conditions.Additionally,the analysis suggested that Qinzhi Zhudan Decoction may have potential new efficacies in the treatment of ataxia and brain glioma.Conclusion:This study elucidates the mechanism of Qinzhi Zhudan Decoction in the treatment of brain disorders from the perspective of target engagement,further supports its potential therapeutic value in central nervous system diseases,including ataxia and glioma.
