中医方药整体作用靶标研究模式探讨 被引量:1

摘要 尊重中医方药整体性和结构性的核心特征,是揭示其疗效机制和多靶点协同作用的关键前提。中医方药通过与机体靶标相互作用发挥药效。当前中医方药靶标研究模式主要以成分为中心,不能体现其整体性和结构性,也不足以阐释中医方药的疗效和作用机制。近年来,基于高通量质谱的配体免修饰检测技术不断发展,为中医方药整体作用靶标的鉴定和亲和力检测研究提供了强大支持。在中医整体观指导下,提出中医方药整体作用靶标研究模式,其包括“以方(药)测靶”“靶标关联”“由标知效”“临床验效”和“传承创新”五大模块:通过基于质谱的配体免修饰检测实验对中医方药作用的靶点蛋白质(即“靶”)进行高通量发现、鉴定和亲和力检测;运用生物信息学解析中医方药作用下产生的生物大数据,遴选与中医方药作用相关的功效分子(即“标”),基于信号传导理论将“靶”与“标”相关联,精确解析方药调控疾病的功效分子;全面构建中医方药治疗疾病的作用网络并挖掘其潜在新功效;进一步回归临床,验证基础研究得出的中医方药创新功效,实现其精准临床定位;最后,对中医方药创新功效开展深入研究,阐释其科学内涵、临床定位和作用机制,促进中医方药理论的传承创新。 Adhering to the core characteristics of traditional Chinese medicine(TCM)prescriptions,including their integrity and structure,is a key prerequisite for revealing their therapeutic mechanisms and multi-target synergistic effects.TCM prescriptions exert medicinal effects by interacting with the target proteins.However,the current research model mainly focuses on individual components,which cannot reflect prescription integrity and is thus insufficient to explain the therapeutic effects and mechanisms of TCM prescriptions.In recent years,the development of unmodified ligand detection technology based on high-throughput mass spectrometry has provided strong support for the identification and affinity detection of the overall action targets of TCM prescriptions.Under the guidance of the holistic view of TCM,this paper proposes a research model for the overall action targets of TCM prescriptions and their components.This model comprises five modules:target identification based on TCM prescriptions,target-effector association,revealing TCM efficacy through effectors,efficacy verification in clinical practice,and inheritance and innovation.The unmodified ligand detection technology basing on mass spectrometry is used for the discovery,identification,and affinity detection of target proteins in a high-throughput way.Then,bioinformatics is employed to analyze the biological big data generated under TCM prescription treatment and select the functional molecules related to the action of TCM prescriptions.According to the theory of signal transduction,targets are associated with functional molecules,and the functional molecules of TCM prescriptions in regulating diseases are precisely analyzed.This could help comprehensively construct the functional network of TCM prescriptions in treating diseases and explore their potential new effects.Further,clinical practice is employed to verify the innovative efficacy of TCM prescriptions that is obtained from the experiments,and thus the precise clinical positioning is achieved.In summary,the in-depth research on the innovative efficacy,scientific concepts,precise clinical position,and therapeutic mechanisms of TCM prescriptions could promote the inheritance and innovation of the theory of TCM prescriptions.
