摘要
目的:筛选已授权的调治冠心病心绞痛的中药专利复方作为优选复方,挖掘其用药及配伍规律,以期为中药新药的研制开发及中药专利的保护提供前期参考。方法:检索国家知识产权局中国专利公布网站中已授权的调治冠心病心绞痛的中药专利复方数据。使用Excel 2016进行数据整理、频数统计;运用IBM SPSS Modeler 18.0与SPSS 25.0分别进行Apriori关联规则分析和系统聚类分析;通过Cytoscape 3.10构建核心中药复杂网络。结果:共纳入优选中药专利复方215项,涉及药物275味,以活血化瘀药和补益药为主。常用的中药为丹参(113次,占52.56%)、川芎(84次,占39.07%)、三七(73次,占33.95%)、黄芪(61次,占28.37%)等;常用的对药为川芎-赤芍(37次,占17.21%)、丹参-葛根(33次,占15.35%)、川芎-当归(32次,占14.88%),丹参-麦冬(27次,占12.56%)、丹参-山楂(24次,占11.16%)等;常用的角药包括丹参-红花-川芎(32次,占14.88%)、川芎-葛根-丹参(27次,占12.56%)、红花-赤芍-川芎(27次,占12.56%)、丹参-赤芍-川芎(27次,占12.56%)、黄芪-当归-川芎(23次,占10.70%)等。高置信度关联规则包括“葛根-川芎→丹参”“赤芍-丹参→川芎”“葛根→丹参”等;核心药物为丹参、川芎、黄芪、三七、红花、地龙、赤芍、当归等。结论:优选中药专利复方调治冠心病心绞痛以益气活血化瘀为主要治法,兼顾冠心病“热结血脉”的过程,适当配伍苦寒药以清瘀热。
Objective:To explore the medication and compatibility rules of the authorized traditional Chinese medicine patent compound formula for treating angina pectoris of coronary heart disease as the preferred compound prescriptions,providing preliminary references for the research and development of traditional Chinese medicine new drugs and the protection of traditional Chinese medicine patents.Methods:All data of authorized traditional Chinese medicine patent compound formula for treating angina pectoris of coronary heart disease were collected from the China Patent Publication Website of the National Intellectual Property Administration.The data organization and frequency statistics were performed by Excel 2016.Apriori association rule analysis and systematic clustering analysis were conducted respectively using IBM SPSS Modeler 18.0 and SPSS 25.0,and the complex network of core traditional Chinese medicine was constructed by Cytoscape 3.10.Results:A total of 215 preferred traditional Chinese medicine patented compound prescriptions were included,involving 275 kinds of drugs,mainly invigorating blood and dissolving stasis drugs,supplement and boost drugs.The commonly used traditional Chinese medicine included Salvia miltiorrhiza(113 times,accounting for 52.56%),Ligusticum Chuanxiong(84 times,accounting for 39.07%),Notoginseng(73 times,accounting for 33.95%),Astragalus membranaceus(61 times,accounting for 28.37%).The commonly used antidrugs were Ligusticum Chuanxiong-Red Peony Root(37 times,accounting for 17.21%),Salvia miltiorrhiza-Pueraria lobata(33 times,accounting for 15.35%),Ligusticum Chuanxiong-Angelica Sinensis(32 times,accounting for 14.88%),Salvia miltiorrhiza-Ophiopogon japonicus(27 times,accounting for 12.56%),Salvia miltiorrhiza-Hawthorn(24 times,accounting for 11.16%).Commonly used horn traditional Chinese medicine included Salvia miltiorrhiza-Safflower-Ligusticum Chuanxiong(32 times,accounting for 14.88%),Ligusticum Chuanxiong-Pueraria lobata-Salvia miltiorrhiza(27 times,accounting for 12.56%),Safflower-Red Peony Root-Ligusticum Chuanxiong(27 times,accounting for 12.56%),Salvia miltiorrhiza-Red Peony Root-Ligusticum Chuanxiong(27 times,accounting for 12.56%),Astragalus membranaceus-Angelica sinensis-Ligusticum Chuanxiong(23 times,accounting for 10.70%).High-confidence association rules included"Pueraria lobata-Ligusticum Chuanxiong→Salvia miltiorrhiza""Red Peony Root-Salvia miltiorrhiza→Ligusticum Chuanxiong""Pueraria lobata→Salvia miltiorrhiza".The core ingredients included Salvia miltiorrhiza,Ligusticum Chuanxiong,Astragalus membranaceus,Notoginseng,Safflower,Earthworm,Red Peony Root,Angelica sinensis.Conclusion:The preferred traditional Chinese medicine patented compound formula for treating coronary heart disease of angina pectoris mainly focused on boosting Qi,invigorating blood and dissolving stasis.It also taked into account the process of"heat accumulation in the blood vessels"of coronary heart disease with bitter and cold herbs to clear the stasis and heat.
