摘要
目的 研究湖北海棠叶中总黄酮的提取分离工艺。方法 以根皮苷为对照,采用正交试验法,通过考察乙醇浓度、提取时间、提取温度、料液比等四个影响因素对有效成分提取的影响,从而优选湖北海棠叶总黄酮提取的最佳条件。所得总黄酮粗品经AB-8大孔吸附树脂、聚酰胺树脂、Sephadex LH-20凝胶树脂三种分离介质分离纯化,采用HPLC法进行检测。结果 提取条件:乙醇浓度为70%、提取温度为50℃、料液比为1∶30、提取时间为2 h时所得总黄酮含量最高;三种分离介质中,Sephadex LH-20凝胶树脂为最佳分离介质,分离效果好纯度高。结论 研究采取的提取分离纯化技术为湖北海棠叶中黄酮的综合利用奠定了基础。
Objective to study the technology on extracting and separating of flavones from leaves of Malus hupehensis.Method In order to extract and separate the flavones from Malus hupehensis,the orthogonal experiments were used to investigate the effects of ethanol concentration,extraction time,extraction temperature and solid-liquid ratio on the extraction.The optimal conditions for the extraction were selected.The total flavonoids were separated and purified by different medium,polyamide resin、AB-8 macro porous adsorption resins and Sephadex LH-20 were used.The content of flavones were determined by HPLC and using Phloridzin for reference substance.Result It shows that the optimal concentration of ethanol for extraction is 70%,the temperature is 50℃,the ratio of material to solvent is 1:30 and the extracting time is 2 hours,respectively.As for purification,among the three medium,the most effective medium is Sephadex LH-20.Conclusion This study paves the way for the utilization of flavones from Malus hupehensis.
