摘要
目的采用HPLC法测定不同产地的沙棘成分,并建立沙棘的二维相关红外光谱分析方法。方法采用Agilent Eclipse Plus C_(18)色谱柱(250 mm×4.6 mm,5μm),测定沙棘中原儿茶酸、芦丁、槲皮素、山奈酚、异鼠李素等成分的含量;采用傅里叶变换红外光谱技术对11批沙棘样品进行分析,通过对红外光谱数据的解析、聚类分析、主成分分析、偏最小二乘法-判别等方法,研究不同地区沙棘药材的分类情况。结果11批样品中5种成分总含量为0.63~2.12 mg·g^(-1);二维相关红外光谱显示:不同产地沙棘的峰形、峰位、吸收强度有明显差异,化学计量学分析从不同层面将不同产地的样品进行分类。结论所用方法均能区分不同产地沙棘并进行分类,结果可靠,可用于沙棘的成分测定与快速鉴别。
OBJECTIVE To detect the components of Hippophae rhamnoides from different regions by HPLC,and to establish a two-dimensional correlation infrared spectroscopy method.METHODSSThe content of procatechuic acid,rutin,quercetin,kaempferol and isorhamnetin in H.rhamnoides were determined by HPLC using Agilent Eclipse Plus Cis column(250 mm×4.6 mm,5μm).11 batches of H.rhamnoides samples were analyzed by Fourier transform infrared spectroscopy,and the classification of H.rhamnoides in different regions was studied using infrared spectral data analysis,cluster analysis,principal component analysis and least-square discriminant analysis.RESULTSSThe total content of 5 components in 11 batches of samples was 0.63-2.12 mg·g^(-1).The two-dimensional correlation infrared spectrum showed that the peak shapes,peak positions and absorption intensities of H.rhamnoides from different places were obviously different.The samples from different places were classified from different levels by stoichiometric analysis.CONCLUSION Both methods can be used to distinguish and classify H.rhamnoides from different places.The results are reliable and can be used for the composition determination and rapid identification of H.rhamnoides.
