摘要
为优化超声法联合水蒸气蒸馏法提取温郁金中的挥发油的最佳工艺条件,以温郁金挥发油得率为考察指标,采用单因素试验考察液料比、超声时间、超声功率、提取时间对温郁金挥发油得率的影响,再应用Box-Behnken响应面法优化挥发油提取工艺条件,通过对比试验考察了水蒸气蒸馏法和超声辅助水蒸气蒸馏法所得郁金挥发油得率及化学成分的差异,并通过体外抗氧化试验考察了最优条件下所得温郁金挥发油对DPPH自由基、羟基自由基和ABTS^(+)自由基的清除活性。结果表明,采用超声法联合水蒸气蒸馏法提取挥发油最佳提取工艺为:液料比为18:1(mL/g)、超声时间为20 min、超声功率为253 W、提取时间为112 min,此时温郁金挥发油得率为3.021%;经对比试验可知,超声法联合水蒸气蒸馏法比传统水蒸气蒸馏法所得温郁金挥发油得率提高了40.0%,经GC-MS法分析以上两种方法所得温郁金挥发油化学成分分别有49个、38个。最佳工艺条件下所得温郁金挥发油清除ABTS^(+)自由基、羟自由基和DPPH自由基的IC_(50)分别为0.373、18.23、0.706 mg/mL,说明超声法联合水蒸气蒸馏法所得温郁金挥发油有较强的抗氧化活性。
In order to optimize the optimum conditions for the extraction of volatile oil from Curcumae Radix by ultrasonic method combined with steam distillation,the yield of volatile oil from Curcumae Radix was used as the index.The effects of liquid-to-materi-al ratio,ultrasonic time,ultrasonic power and extraction time on the yield of volatile oil from Curcumae Radix were investigated by single factor test.The Box-Behnken response surface method was used to optimize the extraction conditions of volatile oil.The differ-ences in the yield and chemical composition of volatile oil obtained by steam distillation and ultrasound-assisted steam distillation were investigated by comparative experiments.The antioxidant activity of the volatile oil obtained under the optimal conditions on DPPH free radical,hydroxyl radical and ABTS+free radical scavenging activity was investigated by in vitro antioxidant test.The re-sults showed that the optimum extraction process of volatile oil by ultrasonic method combined with steam distillation was as follows:the ratio of liquid to material was 18∶1(mL/g),the ultrasonic time was 20 min,the ultrasonic power was 253 W,and the extraction time was 112 min.The yield of volatile oil from Curcumae Radix was 3.021%.The comparison test showed that the yield of volatile oil from Curcumae Radix obtained by ultrasonic method combined with steam distillation was 40.0%higher than that obtained by steam distillation.The GC-MS analysis showed that there were 49 and 38 chemical components of volatile oil from Curcumae Radix obtained by the above two methods.The IC50 of scavenging ABTS+free radical,hydroxyl radical and DPPH free radical of the vola-tile oil obtained under the optimum conditions were 0.373,18.23 and 0.706 mg/mL,respectively,indicating that the volatile oil ob-tained by ultrasonic method combined with steam distillation had strong antioxidant activity.
