蕨麻组培苗消毒条件及组织培养条件的筛选研究

摘要 以蕨麻茎尖为外植体材料,筛选不同消毒体系的灭菌效果及不同激素组合对愈伤组织分化诱导的影响。结果表明,最优消毒方法为75%酒精60s+0.1%HgCl_(2)5min,且对比发现NaClO的灭菌效果不如HgCl_(2);愈伤组织诱导时,最佳培养基的激素组合为在MS培养基中添加1.0mg/L的6-BA 0.6mg/L的NAA,此条件下愈伤组织分化率高,且对比发现,NAA对蕨麻的分化诱导影响强于IBA,而IBA的分化诱导能力又强于2,4-D。 In this study,the stem tip of Potentilla Anserina L.was used as explant material to screen the sterilization effect of different disinfection systems and the effect of different hormone combinations on callus differentiation induction.The results go as the following:the optimal disinfection method is 75%alcohol 60s+0.1%HgCl_(2)5min,and the sterilization eff ect of NaClO was not as good as that of HgCl_(2);in callus induction,the hormone combination of the best medium is to add 1.0mg/L 6-BA 0.6mg/L NAA to the MS medium with high callus diff erentiation rate under this condition;the eff ect of NAA on the diff erentiation induction of Potentilla Anserina L.is stronger than that of IBA,and the diff erentiation induction ability of IBA is stronger than that of 2,4-D.
