摘要
采用超声辅助乙醇-硫酸铵双水相法提取紫苏叶总黄酮。以总黄酮得率为指标,在单因素试验的基础上通过正交试验优化提取工艺,并对其抗氧化活性进行考察。结果表明:最佳提取工艺为硫酸铵质量浓度0.30 g/mL、液料比35∶1(mL/g)、提取时间30 min、乙醇体积分数60%,在此条件下紫苏叶总黄酮得率达15.497%。抗氧化活性试验结果表明,紫苏叶总黄酮对DPPH自由基和ABTS自由基的IC50分别为0.276、0.136 mg/mL,表明紫苏叶总黄酮具有较好的抗氧化活性。
The total flavonoids from Perilla frutescens L.leaves were extracted by ultrasonic-assisted ethanol-ammonium sulfate aqueous two-phase method.Taking the yield of total flavonoids as index,the extraction process was optimized by orthogonal experiment on the basis of single factor experiments,and its antioxidant activity was investigated.The results showed that the optimal extraction process was ammonium sulfate mass concentration 0.30 g/mL,liquid-solid ratio 35∶1(mL/g),extraction time 30 min,and ethanol volume fraction 60%.Under these conditions,the yield of total flavonoids from Perilla frutescens L.leaves reached 15.497%.The antioxidant activity test showed that IC 50 on DPPH and ABTS free radicals were 0.276 mg/mL and 0.136 mg/mL,respectively,indicating that the total flavonoids from Perilla frutescens L.leaves had good antioxidant activity.
