摘要
采用低共熔溶剂提取“温185”核桃分心木中总黄酮。在单因素试验的基础上,以核桃分心木总黄酮提取量为指标,通过响应面试验优化提取工艺,并对其抗氧化活性进行研究。结果表明:最佳提取工艺为低共熔溶剂氢键受体与供体摩尔比3∶1、含水率15%、超声时间35 min、超声温度70℃,在此条件下总黄酮提取量平均值为16.76 mg/g。抗氧化试验结果表明,与传统水提法比较,低共熔溶剂组的总黄酮提取物具有良好的抗氧化活性,当质量浓度为1.0 mg/mL时,其对DPPH、ABTS自由基的清除率可达85.33%、78.23%。
Total flavonoids ofWen 185walnut Diaphragma juglandis fructus(DJF)were extracted using deep eutectic solvent.Taking the extraction amount of total flavonoids in DJF as the index based on the single factor experiments,the extraction process of total flavonoids was optimized through the response surface experiment,and its antioxidant activity was studied.The results showed that the optimal extraction process was molar ratio of hydrogen bond acceptor to donor of deep eutectic solvent 3∶1,water content 15%,ultrasonic time 35 min,and ultrasonic temperature 70℃.Under these conditions,the average extraction amount of total flavonoids was 16.76 mg/g.The antioxidant experiment results showed that compared with the traditional water extraction method,the total flavonoids extract of the deep eutectic solvent group had good antioxidant activity.When the mass concentration was 1.0 mg/mL,its scavenging rates of DPPH and ABTS free radicals could reach 85.33%and 78.23%,respectively.
