外耳道胆脂瘤动物模型的构建 被引量:2

An Animal Model of External Auditory Canal Cholesteatoma

摘要 目的 探讨外耳道胆脂瘤动物模型简易可靠的构建方法。方法 选取沙鼠和C57BL/6小鼠各10只共40耳,采用外耳道结扎法构建外耳道胆脂瘤动物模型,术后6、8、10、12、16周通过肉眼、组织病理学及显微CT扫描评估胆脂瘤形成情况。结果 沙鼠及C57BL/6小鼠外耳道结扎后,耳部胆脂瘤自外耳道向中耳方向扩展,组织病理学和影像学可以相对早期判定胆脂瘤是否形成;外耳道胆脂瘤成模理想时间为术后3个月,术后4个月常见到中耳受累。结论 沙鼠和C57BL/6小鼠通过外耳道结扎法均可以成功构建外耳道胆脂瘤动物模型,该建模方法简便易行、成模率高,值得在未来相关基础研究中运用。 Objective To report a simple and reliable method for establishing an animal model of external auditory canal cholesteatoma.Method Mongolian gerbils(n=10) and C57BL/6 mice(n=10)(total 40 ears) were selected.The external auditory canal was ligated and the formation of cholesteatoma was visually evaluated and by histopathology and micro-CT scanning at 6,8,10,12 and 16 weeks postoperatively.Results Following ligation of the external ear canal,cholesteatoma formed at about 3 months and expanded from the external ear canal to the middle ear at roughly 4months,evidenced by histopathology and imaging at relatively early stages.Conclusion Animal models of external auditory canal cholesteatoma can be achieved by ligation of the external auditory canal,which is simple and easy with a high rate of success,and worthy of adoption in research.
