氨苯砜胶体金免疫层析试纸条研制及其在牛尿液样本检测中的应用 被引量:2

摘要 为了检测牛尿液中的氨苯砜残留量,依据胶体金免疫层析原理研制了氨苯砜快速检测试纸条。使用氨苯砜-鸡卵白蛋白偶联物作为人工抗原免疫新西兰大白兔获得多克隆抗体,使用胶体金标记多克隆抗体,试纸条的检测线包被氨苯砜-牛血清白蛋白(BSA)偶联物,质控线包被羊抗兔IgG抗体。用试纸条对牛尿中的氨苯砜残留量进行检测,其检测限为5μg/L,检测时间10 min,特异性较好,与氨苯砜相似的几种药物均无交叉反应。本试验研制的试纸条可以快速检测牛尿液中氨苯砜残留量,实时监测牛在饲养过程中是否使用氨苯砜,为控制氨苯砜残留提供了有效监管手段。 In order to detect the residue of dapsone in bovine urine,a rapid test strip for dapsone detection was developed,based on colloi-dal gold immunochromatography.New Zealand white rabbits were used and immunized with dapsone-OVA conjugate as an artificial antigen to obtain polyclonal antibodies.The polyclonal antibodies were labeled with colloidal gold.The test line of the strip was coated with dapsone BSA conjugate,and the quality line was coated with goat anti-rabbit IgG.The limit of detection was 5μg/kg,in 10 minutes,when the residue of dapsone in cow urine was determined.The results showed that the test strip had no cross reaction with some drugs similar to dapsone,that it could quickly detect the residue of dapsone in cow urine,and that it could serve as an effective supervision means for controlling the residue of dapsone and could monitor the use of dapsone in cattle feeding.
