摘要
该研究建立了同位素内标液相色谱-串联质谱法检测白酒、啤酒、葡萄酒与黄酒中γ-氨基丁酸(GABA)的方法。酒样稀释后加入内标物γ-氨基丁酸-d6,在Acclaim Trinity P1色谱柱上,以乙腈(含0.1%甲酸)+10 mmol/L乙酸铵溶液(1∶1,V/V)为流动相,采用电喷雾正离子多反应监测(MRM)模式采集数据,以保留时间定性,内标法定量。结果表明,在10~500μg/L质量浓度范围内,标准曲线的线性关系良好,相关系数R2为0.999 9。白酒、啤酒、葡萄酒、黄酒中γ-氨基丁酸检出限为0.02~1.64 mg/L,定量限为0.05~5.46 mg/L。精密度实验结果相对标准偏差(RSD)为0.86%~2.04%,加标回收率为88.8%~113.1%。该方法操作简单、快捷、灵敏度高,并具有良好的精密度与准确度,适用于酒类产品中γ-氨基丁酸的测定。
A method for determination ofγ-aminobutyric acid(GABA)in Baijiu,beer,wine and Huangjiu by liquid chromatography-tandem mass spectrometry using isotope internal standard was established.The diluted sample was spiked withγ-aminobutyric acid-d6,separated by Acclaim Trinity P1 column,with the acetonitrile containing 0.1%formic acid and 10 mmol/L ammonium acetate aqueous solution(1∶1,V/V)as mobile phase,and the GABA was detected by electrospray ionization in positive ion mode with multiple reaction monitoring(MRM).The samples were qualitatively determined with retention time and quantified by the internal standard method.Results showed that GABA presented good liner relationship in the range of 10-500μg/L,and the correlation coefficient R2 was 0.9999.The detection limit of GABA in Baijiu,beer,wine and Huangjiu was 0.02-1.64 mg/L,and the quantitative limit was 0.05-5.46 mg/L,respectively.The relative standard deviation(RSD)of precision tests was 0.86%-2.04%,and the spiked recoveries ranged from 88.8%to 113.1%.The method was simple,rapid,with high sensitivity and satisfactory precision and accuracy,which was suitable for the determination ofγ-aminobutyric acid in alcoholic beverage.
