摘要
【目的】探明蛹虫草芡实保健饮料的制备及其对果蝇生殖力和寿命的影响,为蛹虫草大众食品和饮品等保健产品的研制提供参考。【方法】以蛹虫草和芡实为材料,采用单因素和正交试验方法,研究蛹虫草芡实保健饮料制备的最佳配比及其对果蝇生殖力和寿命的影响。【结果】蛹虫草浸提液和芡实滤液体积比为3∶7,蜂蜜、白砂糖和柠檬酸添加量分别为10%、8%和0.10%时,蛹虫草芡实保健饮料的颜色淡黄、清亮,酸甜爽口,具有典型的蛹虫草香味和芡实清香,其pH为4.5~4.8,细菌总数≤100 CFU/mL,大肠杆菌≤3 MPN/mL,符合国家食品生产卫生标准。CK(对照培养基),F_(1)和F_(2)的成蛹数、成虫数分别为65.98只、59.13只和64.43只、57.02只。10%饮料饲喂果蝇,F_(1)(76.44只、68.08只)和F_(2)(72.89只、64.16只)的成蛹数、成虫数分别较CK提高15.85%、15.14%和13.13%、12.52%;15%饮料饲喂果蝇,F_(1)(79.98只、71.62只)和F_(2)(77.02只、70.01只)的成蛹数、成虫数分别较CK提高21.22%、21.12%和19.54%、22.78%。CK雌(♀)果蝇和雄(♂)果蝇半数死亡时间、平均寿命和最高寿命分别为55.13 d、60.43 d、74.26 d和55.81 d、60.32 d、78.56 d。10%饮料饲喂雌果蝇(61.18 d、65.16 d、82.63 d)和雄果蝇(61.86 d、66.79 d、89.02 d),其半数死亡时间、平均寿命、最高寿命分别较CK提高10.97%、7.82%、11.27%和10.84%、10.73%、13.31%;15%饮料饲喂雌果蝇(62.85 d、68.35 d、85.90 d)和雄果蝇(65.25 d、69.11 d、91.82 d),分别较CK提高14.01%、13.11%、15.67%和16.91%、14.57%、16.88%。【结论】蛹虫草芡实保健饮料的最佳配方为蛹虫草浸提液和芡实滤液体积比3∶7,同时添加10%蜂蜜、8%白砂糖和0.10%柠檬酸;适宜浓度(10%、15%)的饮料饲喂果蝇能明显提高其生殖力和寿命。
【Objective】The preparation of Cordyceps militaris and Gordon euryale health drink and its effect on the fecundity and lifespan of Drosophila are explored to provide a reference for the development of C.militaris popular food and drink and other health products.【Method】Taking C.militaris and G.euryale as materials,the optimal proportion of C.militaris and G.euryale health drink and its effects on the fecundity and lifespan of Drosophila are studied by single factor and orthogonal test.【Result】When the volume ratio of C.militaris extract to G.euryale filtrate is 3∶7,and the addition of honey,sugar and citric acid is 10%,8% and 0.10% respectively,the health drink of C.militaris and G.euryale is light yellow,clear,sweet and sour,with a typical smell of C.militaris and the scent of G.Euryale.Its pH is 4.5~4.8,and the total number of bacteria is less than 100 CFU/mL,and Escherichia coli is less than 3 MPN/mL,which meet the national food production hygienic standard.The number of pupae and adults of CK,F_(1) and F_(2) is 65.98,59.13,and 64.43,57.02,respectively.When Drosophila is fed with 10% drink,the number of pupae and adults of F_(1)(76.44,68.08) and F_(2)(72.89,64.16) are 15.85%,15.14%,and 13.13%,12.52% higher than CK.When Drosophila is fed with 15% drink,the number of pupae and adults of F_(1)(79.98,71.62) and F_(2)(77.02,70.01) are 21.22%,21.12%,and 19.54%,22.78% higher than CK.The median lethal time,average lifespan and maximum lifespan of female Drosophila and male Drosophila is 55.13 d,60.43 d,74.26 d,and 55.81 d,60.32 d,78.56 d,respectively.The median lethal time,average lifespan and maximum lifespan of female Drosophila(61.18 d、65.16 d and 82.63 d) and male Drosophila(61.86 d、66.79 d、89.02 d)fed with 10% drink is respectively increased by 10.97%,7.82%,11.27% and 10.84%,10.73%,13.31% compared with CK.The median lethal time,average lifespan and maximum lifespan of female Drosophila(62.85 d,68.35 d and 85.90 d) and male Drosophila(65.25 d,69.11 d and 91.82 d)fed with 15% drink is respectively increased by 14.01%,13.11%,15.67% and 16.91%,14.57%,16.88% compared with CK.【Conclusion】The optimum formula of C.militaris and G.euryale health drink includes 3∶7 of the volume ratio of C.militaris extract to G.euryale filtrate,and adding 10% honey,8% sugar and 0.10% citric acid at the same time;The suitable concentration(10%,15%) of the drink can significantly improve the fecundity and lifespan of Drosophila.
