经典名方当归建中汤HPLC指纹图谱及含量测定方法研究 被引量:2

摘要 目的建立经典名方当归建中汤(DJD)HPLC指纹图谱及主要成分含量测定方法。方法按照传统制法制备DJD基准样品,利用HPLC法建立基准样品的指纹图谱,进行相似度评价及色谱峰归属,选择效应关联成分阿魏酸、芍药苷、甘草苷、肉桂酸、桂皮醛作为指标成分,建立基准样品的含量测定方法。结果15批DJD基准样品指纹图谱的相似度均大于0.9,确定了28个共有峰;5个指标成分含量测定方法学考察均符合2020年版《中国药典》规定,DJD基准样品中阿魏酸含量为0.028%~0.034%,芍药苷含量为1.498%~2.143%、甘草苷含量为0.082%~0.318%、肉桂酸含量为0.012%~0.025%、桂皮醛含量为0.002%~0.012%。结论建立的方法准确、稳定、重复性好,为DJD基准样品质量控制及复方制剂的开发提供了参考。 Objective To establish HPLC fingerprint of the substance benchmarks of the ancient classical prescriptions of Danggui Jianzhong Decoction(DJD)and the content determination of main index components.Methods The DJD reference sample is prepared according to the traditional preparation method,the fingerprint of the reference sample is established by the HPLC.The attribution of the chromatographic peaks has been determined and the similarity is evaluated.At the same time,the methods are established for determining the content of ferulic acid,paeoniflorin,liquiritin,cinnamic acid and cinnamaldehyde by HPLC and the contents are determined respectively.Results In the 15 batches of the reference samples of DJD,the similarity of fingerprints are higher than 0.9.There are 28 common peaks in the fingerprints of DJD.These five quality control component content determination methodological investigations all met the requirements of the Chinese Pharmacopoeia(2020).The content of ferulic acid in the corresponding objects is 0.028%~0.034%,paeoniflorin is 1.498%~2.143%,liquiritin is 0.082%~0.318%,cinnamic acid is 0.012%~0.025%,cinnamaldehyde is 0.002%~0.012%.Conclusion This accurate,stable and reproducible method can provide reference for the development of DJD reference sample quality standards and compound preparation.
