摘要
目的筛选木蝴蝶总黄酮的最优提取工艺。方法以木蝴蝶苷B为对照品,采用紫外-可见分光光度法,以木蝴蝶总黄酮含量为评价指标,以提取方法、提取溶剂种类、提取溶剂体积分数、提取温度、提取时间、料液比为考察因素,优选最佳提取工艺,并进行验证。结果最佳提取工艺为采用回流提取法,以75%乙醇为提取溶剂,料液比为1∶20(m/V),提取温度为60℃,提取1.5 h;木蝴蝶总黄酮平均含量为2.23 mg/g,RSD为1.49(n=3)。结论该提取工艺操作简单,结果可靠,数据准确,可为木蝴蝶黄酮类化合物的开发利用提供参考。
Objective To optimize the extraction process of total flavonoids from Oroxyli Semen.Methods The Oroxin B was taken as the reference,the ultraviolet-visible(UV-Vis)spectrophotometry method was adopted to optimize and verify the extraction process with the total flavonoids content of the Oroxyli Semen as the evaluation index and the extraction method,type of extraction solvent,volume fraction of extraction solvent extraction temperature,extraction time and solid/liquid ratio as investigation factors.Results The optimal extraction process was reflux extraction,75%ethanol as extraction solvent,solid/liquid ratio of 1∶20(m/V),extraction temperature of 60℃and extraction time of 1.5 h.The average content of total flavonoids of the Oroxyli Semen was 2.23 mg/g with an RSD of 1.49(n=3).Conclusion The extraction process is simple,reliable and accurate,which can provide a reference for the development and utilization of flavonoids in Oroxyli Semen.
