降压宝蓝片对自发性高血压大鼠血管重构的影响 被引量:4

Effect of Jiangya Baolan Tablet on Vascular Remodeling in Spontaneously Hypertensive Rats

摘要 目的:研究降压宝蓝片对自发性高血压大鼠(spontaneously hypertensive rats,SHR)血管重构(vascular remodeling,VR)及肾素-血管紧张素系统(renin-angiotensin system,RAS)/激肽释放酶-激肽系统(kallikrein-kinin system,KKS)平衡的影响。方法:30只雄性SHR随机分为模型组(0.5%羧甲基纤维素钠,carboxymethyl cellulose,CMC)、降压宝蓝片组(0.6 g·kg^(-1))、依那普利组(0.02 g·kg^(-1)),另设同龄正常血压的雄性WKY 10只为空白组(0.5%CMC),灌胃给药连续12周。取胸主动脉并观察病理形态改变,测定中膜厚度(media thickness,MT)、中膜胶原蛋白容积分数(collagen volume fraction,CVF)和中膜弹力纤维容积分数(elastin volume fraction,EVF),腹主动脉取血,Elisa法测定血清血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)和缓激肽(bradykinin,BK)含量,微板法测定血清一氧化氮(nitric oxide,NO)含量。结果:与空白组比较,模型组大鼠胸主动脉MT、CVF明显增加(P<0.01),EVF及血清BK含量显著降低(P<0.05),说明降压宝蓝片能显著降低主动脉MT和CVF(P<0.01),明显升高主动脉EVF及血清BK、NO含量(P<0.05)。结论:降压宝蓝片具有改善高血压血管重构的作用,部分作用机制与调节RAS/KKS平衡有关。 Objective:To study the effects of Jiangya Baolan Tablet(JYBLP)on vascular remodeling(VR)and the balance of renin-angiotensin system(RAS)/kallikinase kinin system(KKS)in spontaneously hypertensive rats.Methods:30 male SHR were randomly divided into model group(0.5%CMC),JYBLP group(0.6 g·kg^(-1)),enalapril group(0.02 g·kg^(-1)),and 10 Wistar-kyoto(WKY)rats were used as blank group(0.5%CMC).After12 weeks treatment,Thoracic aorta was taken to observe pathological changes,media thickness(MT),collagen volume fraction(CVF)and elastic fiber volume fraction(EVF).Blood was taken from abdominal aorta,the levels of angiotensinⅡ(AngⅡ)and bradykinin(BK)in serum were detected by enzyme-linked immunosorbent assay(ELISA),nitric oxide(NO)was observed by microplate method.Results:Compared with blank group,the contents of MT and CVF in thoracic aorta were increased significantly(P<0.01),EVF in thoracic aorta and BK in serum were decreased significantly(P<0.05).JYBLP could significantly decrease MT,CVF(P<0.01)in thoracic aorta,significantly increase the contents of EVF in thoracic aorta and BK,NO in serum(P<0.05).Conclusion:JYBLP could improve vascular remodeling in hypertension,the mechanism is partly related to the regulation of RAS/KKS balance.
