摘要
果葡糖浆(HFCS)因甜度高、流动性好,被广泛应用于甜味剂。现有果葡糖浆的分析方法大都受到分析成本高、周期长等因素的制约,而且难以兼备准确性和高效性。文章采用拉曼光谱结合角度转换建立了果葡糖浆的快速分析方法。首先以市售果葡糖浆为建模样本母液,配制不同果糖含量的分析样本,采集拉曼光谱;经荧光褪色差分法(FBDA)基线校正和一阶滤波求导平滑后,截取特征波段;通过子空间角度转换算法计算得到各样本与参比样本光谱的夹角值EE,建立样本中果糖含量和EE值的关联模型,模型相关系数R达0.9991。利用关联模型,分析市售果葡糖浆中果糖含量。平行分析各样品5次,计算相对标准偏差(RSD)在0.60%-2.32%之间,与分光光度法测定结果比对,相对误差在1.89%-6.77%之间。结果表明,拉曼光谱与角度转换的化学计量学方法联合可用于市售果葡糖浆中果糖的快速定量分析,该方法精密度高、准确性好、分析效率高,可用于果葡糖浆产品的质量监控。
High fructose corn syrup(HFCS)is widely used as a sweetener because of its high sweetness and good fluidity.However,most of the existing analytical methods of HFCS are restricted by high analysis cost and long period,and it is difficult to have both accuracy and efficiency.Raman spectroscopy combined with angle conversion is used to establish a fast analytical method for HFCS.Firstly,a series of analytical samples with different fructose content are prepared with commercial fructose syrup as the mother liquid of modeling sample,and Raman spectra are collected;after FBDA baseline correction and first-order filter derivation smoothing,the characteristic bands are intercepted;by using the subspace angle conversion algorithm,the included angle value EE of each sample and the reference sample is calculated,and the correlation model of fructose content and EE value in the sample is established,and the correlation coefficient R of the model reaches 0.9991.The content of fructose in commercial fructose syrup is analyzed by using correlation model.The relative standard deviations(RSDs)are in the range of 0.60%-2.32%,and the relative errors are in the range of 1.89%-6.77%compared with the results obtained by spectrophotometry.The results show that the combination of Raman spectroscopy and angle conversion stoichiometry could be used for the rapid quantitative analysis of fructose in HFCS.The method has high precision,accuracy and efficiency,and could be used for the quality control of HFCS.
