摘要
建立了HPLC法测定22种中药材中黄曲霉毒素的方法。样品经70%甲醇提取,经免疫亲和柱净化,以InertSustain C 18色谱柱(4.6 mm×250 mm,5μm)分离,梯度洗脱,经荧光检测器检测。结果表明:经方法学验证,4种黄曲霉毒素线性关系良好(R>0.990),检测限(LOD)和定量限(LOQ)分别为0.2~0.5μg/kg和0.4~1.0μg/kg,低、中、高3个添加浓度水平的平均回收率为63.0%~104.2%,相对标准偏差(RSD)为0.5%~1.7%。该方法结果准确且重复性好,适用于22种中药材中黄曲霉毒素的检测。
A method for the determination of aflatoxins in 22 kinds of traditional chinese medicine by high performance liquid chromatography was established.The sample was extracted by 70%methanol,purified by an immunoaffinity column,then,it was separated by InertSustain C 18 chromatographic column(4.6 mm×250 mm,5μm)with gradient elution and detected by fluorescence detector.The results showed that the linear relationship of the four aflatoxins was good(R>0.990),the detection limit(LOD)and quantitative limit(LOQ)were 0.2~0.5μg/kg and 0.4~1.0μg/kg,respectively,the average recoveries were 63.0%~104.2%and the relative standard deviation(RSD)was 0.5%~1.7%.The method is accurate,reproducible and suitable for the determination of aflatoxins in 22 kinds of traditional Chinese Medicine.
