孕鼠肥胖对子代雄鼠早期肝脏脂肪代谢的影响和机制研究 被引量:1

摘要 目的探讨肥胖母鼠对子代雄鼠早期肝脏脂肪代谢的影响以及自噬所起的作用。方法采用随机数字表法将20只雌鼠分为肥胖母鼠(OM)组和普通母鼠(CM)组,每组10只。OM组雌鼠接受高脂饲料(含60%脂肪)喂养,CM组雌鼠接受普通饲料(含4%脂肪)喂养。它们的后代雄鼠被定义为OM子代鼠(OM-O)组、CM子代鼠(CM-O)组。采用油红O染色法观察两组子代雄鼠肝脏形态学的改变,ELISA法检测两组子代雄鼠血清TC、TG、HDL、LDL水平。并进一步采用RT-PCR法和Western blot法检测肝脏组织中自噬相关基因Beclin-1、自噬相关基因3(ATG3)、自噬相关基因5(ATG5)、自噬相关基因12(ATG12)、p62及轻链3B(LC3B)的变化,免疫组化染色法检测肝脏组织中LC3B的含量。结果与CM-O组比较,OM-O组肝脏切片中脂滴形成不明显,肝脏脂滴沉积较轻;两组子代雄鼠血清TC、TG、HDL、LDL水平比较差异均无统计学意义(均P>0.05);与CM-O组比较,OM-O组Beclin-1、LC3B mRNA表达水平均上调,p62 mRNA表达水平下调,差异均有统计学意义(均P<0.05);与CM-O组比较,OM-O组Beclin-1、ATG12、LC3B蛋白表达水平均上调,p62表达水平下调,差异均有统计学意义(均P<0.05);OM-O组肝脏组织中LC3B的积分光密度值为2.43±1.02,高于CM-O组的0.70±0.20,差异有统计学意义(P<0.05)。结论与CM-O组比较,OM-O组早期肝脏变化相对较轻,这种母代肥胖对其子代肝脏脂肪代谢影响可能与自噬在其中发挥了一定的积极作用有关。 Objective To investigate the effect of maternal obesity on the early changes of liver fat metabolism in offspring male rats and related mechanisms.Methods Twenty female rats were randomly divided into obese maternal(OM)group(n=10)and common maternal(CM)group(n=10).The female rats in the OM group were fed with a high-fat diet(containing 60%fat),and the female mice in the CM group were fed with a normal diet(containing 4%fat).Their offspring were defined as obese maternal offspring(OM-O)group and common maternal offspring(CM-O)group.The liver morphological changes of the two groups were observed by oil red O staining,and the serum levels of TC,TG,HDL and LDL in the two groups were detected by ELISA method.The autophagy-related genes Beclin-1,ATG3,ATG5,ATG12,p62 and LC3B were detected by RT-PCR and Western blot methods,and the expression of LC3B in liver was detected by immunohistochemical staining.Results Compared with CM-O group,the formation of lipid droplets in OM-O group was not significant,and the deposition of lipid droplets in liver was lighter,and there were no significant differences in the serum levels of TC,TG,HDL and LDL between the two groups(all P>0.05).Compared with CM-O group,the expression levels of Beclin-1 and LC3B mRNA were up-regulated and the expression level of p62 mRNA was down-regulated in OM-O group(all P<0.05).Compared with CM-O group,the protein expression levels of Beclin-1,ATG12 and LC3B were up-regulated and p62 was down-regulated in OM-O group(all P<0.05).The integral optical density(IOD)of LC3B in liver tissue of OM-O group was 2.43±1.02,which was significantly higher than that of CM-O group(0.70±0.20)(P<0.05).Conclusion Compared with CM-O group,the early liver changes in OM-O group were relatively mild,and the effect of maternal obesity on liver lipid metabolism in their offspring may be related to the autophagy.
