鬼针草总黄酮超声辅助提取工艺优化及抑菌活性研究 被引量:7

Study on the optimization of ultrasonic-assisted extraction technology and antibacterial activity of total flavonoids from Bidens Pilosa L.

摘要 试验旨在优化鬼针草总黄酮超声辅助提取工艺,并对其抑菌作用进行研究,比较超声辅助提取法与传统提取方法的提取效率。试验采用正交设计对超声辅助提取法进行工艺优化;采用琼脂打孔法测定提取物的抑菌活性,采用微量倍比稀释法测定其最低抑菌浓度(MIC)和最小杀菌浓度(MBC)。结果显示:超声辅助回流提取法的提取得率高于其他方法。最佳提取工艺条件为:药材粉末加入10倍量50%乙醇,超声20 min,90℃下回流提取40 min,平均提取得率35.59%,提取物总黄酮平均含量5.54%。正交试验各组的提取物对金黄色葡萄球菌均有抑制作用。高黄酮组的MIC为15.62 g/L,MBC为125 g/L;低黄酮组的MIC为31.25 g/L,MBC为250 g/L。试验确定鬼针草总黄酮提取的最佳工艺和抑菌作用。 The article aimed to optimize the ultrasonic-assisted extraction process of total flavonoids from Bidens bipinnata L. and to study its bacteriostasis. The extraction efficiency was compared between the ultrasonic-assisted extraction method and the traditional extraction method, then the ultrasonic-assisted extraction process was optimized with the method of orthogonal design. The antibacterial activities of the flavonoids extract were determined by agar drilling method, and its MIC and MBC were determined by microdilution method. The result showed that the optimum extraction conditions included: 10 times amount of 50% ethanol for 20 minutes in ultrasonic environment, then refluxed at90 ℃ for 40 minutes. The average yield was 35.59%, and the total flavonoids content was 5.54%. The extracts showed a certain ability to inhibit Staphylococcus aureus. The MIC of the highest content group was 15.62 g/L, and the MBC was125 g/L. The MIC of the lowest content group was 31.25 g/L, and the MBC was 250 g/L. The extract of Bidens bipinnata has inhibitory effect on Staphylococcus aureus. The best extraction technology of total flavonoids and antibacterial effect of Bidens bipinnata L. was established.
