摘要
目的探讨比索洛尔对慢性心力衰竭大鼠心肌细胞功能及MAPK/ERK1/2信号通路的影响。方法将SD大鼠随机分为对照组、模型组、比索洛尔组,每组15只。采用超声心动图检测各组大鼠心功能相关指标;实验结束后,处死大鼠,并对大鼠心肌组织进行HE染色;采用Western blotting检测大鼠心肌组织中MAPK蛋白、p-MAPK蛋白、ERK1/2蛋白、p-ERK1/2蛋白的表达水平。结果3组大鼠心脏功能各指标比较,差异有统计学意义(P<0.01)。模型组大鼠的左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVESD)较对照组变大(P<0.05),舒张末期左心室后壁厚度(LVPWD)、收缩末期左心室后壁厚度(LVPWS)、左心室射血分数(LVEF)较对照组变薄或降低(P<0.05)。比索洛尔组LVEDD、LVESD较模型组变小(P<0.05),LVPWD、LVPWS、LVEF较模型组增厚或升高(P<0.05)。3组大鼠的慢性心力衰竭生化指标比较,差异有统计学意义(P<0.05)。模型组NT-proBNP、Mb、cTnI、CK-MB水平较比索洛尔组和对照组升高(P<0.05),其余相关指标较比索洛尔组和对照组降低(P<0.05)。3组大鼠心肌组织MAPK/ERK信号通路相关蛋白的表达水平比较,差异有统计学意义(P<0.05)。模型组ERK1/2及p-MAPK蛋白的表达水平较对照组升高(P<0.05);比索洛尔组的ERK1/2和p-MAPK蛋白的表达水平较模型组降低(P<0.05)。结论比索洛尔能通过抑制MAPK/ERK1/2信号通路激活,发挥保护心力衰竭大鼠心肌细胞的作用。
Objective To investigate the effects of bisoprolol on the functions of cardiomyocyte and MAPK/ERK1/2 signaling pathway in rats with chronic heart failure.Methods SD rats were randomly divided into three groups:control group,model group and bisoprolol group(n=15).Cardiac function of mice in each group was detected,HE staining was performed on myocardial tissues,and the expression level of related proteins in myocardial tissues was detected.Results The comparison of the indexes of cardiac function in the three groups were statistically significant by analysis of variance(P<0.01).Compared with the control group,LVEDD and LVESD in the model group increased(P<0.05),while LVPWTd,VPWTs and LVEF decreased(P<0.05).Compared with the model group,LVEDD and LVESD in the bisoprolol group decreased(P<0.05),while LVPWTd,LVPWTs and LVEF increased(P<0.05).The comparison of biochemical indexes of chronic heart failure in the three groups was statistically significant by analysis of variance(P<0.05).The levels of NT-proBNP,Mb,cTnI and CK-MB in model group were the highest,followed by bisoprolol group and control group,but the other indexes in model group were the lowest(P<0.05)by pairwise comparison.The comparison of MAPK/ERK signaling pathway-related protein expression in myocardial tissue of the three groups was statistically significant by analysis of variance(P<0.05).Compared with the control group,ERK1/2 and p-MAPK protein levels in the model group increased(P<0.05),while ERK1/2 and p-MAPK protein levels in the bisoprolol group decreased(P<0.05).Conclusion Antisterone can protect cardiac myocytes of rats with heart failure by inhibiting the activation of MAPK/ERK1/2 signaling pathway.
