尼可地尔对急性心肌梗死大鼠心脏功能和冠脉微循环相关因子的影响 被引量:5

Effects of Nicorandil on Cardiac Function and Factors Related to Coronary Microcirculation in Rats with Acute Myocardial Infarction

摘要 目的:观察尼可地尔改善急性心肌梗死(AMI)大鼠心肌损伤、心脏功能和冠脉微循环影响因子的作用。方法:通过结扎冠状动脉前降支建立Wistar大鼠AMI模型(20只),随机分为模型组和尼可地尔组,每组10只,另取同批大鼠10只作为假手术组。尼可地尔组给予尼可地尔灌胃(2.7mg/kg体质量),模型组和假手术组灌胃等体积纯净水,28天后先行心脏彩超观测各组大鼠心脏形态和功能指标左室舒张末期内径(LVEDD)、左室收缩末期内径(LVEDS)、室间隔舒张末期厚度(IVST)、左室射血分数(LVEF),再行腹主动脉采血检测血清肌钙蛋白T(cTnT)、内皮素(ET)、降钙素基因相关肽(CGRP)、6-酮-前列腺素F1α(6-Keto-PGF1α)和血栓烷B2(TXB2)水平,最后摘取心脏,TTC染色测定心肌梗死面积比。结果:与假手术组比较,模型组大鼠LVEDD、LVEDS、cTnT、ET、TXB2、心肌梗死面积比显著增加(P<0.05,P<0.01),IVST、LVEF显著减小(P<0.05,P<0.01);与模型组比较,尼可地尔组大鼠LVEDD、LVEDS、cTnT、ET、TXB2、心肌梗死面积比明显降低(P<0.05,P<0.01),LVEF显著提高(P<0.01)。结论:尼可地尔能改善微循环影响因子,减少心肌梗死面积,保护心脏功能。 Objective:To observe the effects of Nicorandil on myocardial injury and cardiac function as well as coronary circulation related factors in rats with acute myocardial infarction(AMI).Method:Twenty wistar rats were randomly divided into model group and Nicorandil group after the establishment of AMI model(anterior descending branch of coronary artery was ligated),10 rats in each group.In addition,10 rats were assigned to the sham group.Nicorandil group was given by Nicorandil(2.7mg/kg body mass),model group and sham group were given a gavage of equal purified water.After 28 days,cardiac morphological and functional indicators were evaluated by echocardiogram,which included left ventricle end-diastolic diameter(LVEDD),left ventricle end-systolic diameter(LVEDS),inter-ventricular septum end-diastolic thickness(IVST)and left ventricular ejection fraction(LVEF),and the serum cardiac troponin T(cTnT),endothelin(ET),calcitonin gene related peptide(CGRP),6-ketone-prostaglandin F1 alpha(6-Keto-PGF1α)and thromboxane B2(TXB2)were detected,as well as myocardial infarction area ratio were observed by 2,3,5-triphenyltetrazolium chloride staining.Results:Compared with the sham group,LVEDD,LVEDS,cTnT,ET,TXB2 and myocardial infarction area ratio in rats with AMI were significantly increased(P<0.05,P<0.01),IVST and LVEF were significantly reduced(P<0.01,P<0.05).Compared with the model group,LVEDD,LVEDS,cTnT,ET,TXB2 and myocardial infarction area ratio in rats of Nicorandil goup were significantly decreased(P<0.05,P<0.01)as well as LVEF was significantly increased(P<0.01).Conclusion:Nicorandil can improve microcirculation related factors,reduce myocardial infarction size and protect cardiac function.
