摘要
目的建立一测多评法同时测定表虚感冒颗粒(桂枝、白芍、葛根等)中氧化芍药苷、芍药内酯苷、芍药苷、3′-羟基葛根素、葛根素、3′-甲氧基葛根素、染料木苷、肉桂酸、桂皮醛的含有量。方法该药物50%乙醇提取液的分析采用Agilent ZORBAX SB-C(18)色谱柱(250 mm×4.6 mm,5μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量0.9 mL/min;柱温25℃;检测波长230、250、280 nm。以葛根素为内标,计算其他8种成分的相对校正因子,测定含有量。结果9种成分在各自范围内线性关系良好(r≥0.9991),平均加样回收率96.98%~100.08%,RSD 0.75%~1.54%。一测多评法所得结果与外标法接近。结论该方法简便准确,可用于表虚感冒颗粒的质量控制。
AIM To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous content determination of oxypaeoniflorin,albiflorin,paeoniflorin,3′-hydroxy puerarin,puerarin,3′-methoxy puerarin,genistin,cinnamic acid and cinnamaldehyde in Biaoxu Ganmao Granules(Cinnamomi Ramulus,Paeoniae Radix Alba,Puerariae lobatae Radix,etc.).METHODS The analysis of 50%ethanol extract of this drug was performed on a 25℃thermostatic Agilent ZORBAX SB-C18 column(250 mm×4.6 mm,5μm),with the mobile phase comprising of acetonitrile-0.1%formic acid flowing at 0.9 mL/min in a gradient elution manner,and the detection wavelengths were set at 230,250,280 nm.Puerarin was used as an internal standard to calculate the relative correction factors of the other eight constituents,after which the content determination was made.RESULTS Nine constituents showed good linear relationships within their own ranges(r≥0.9991),whose average recoveries were 96.98%-100.08%with the RSDs of 0.75%-1.54%.The results obtained by QAMS approximated those obtained by external standard method.CONCLUSION This simple and accurate method can be used for the quality control of Biaoxu Ganmao Granules.
