摘要
目的建立高效液相色谱-二极管阵列检测器(HPLC-DAD)同时测定鸢都感冒颗粒中绿原酸、葛根素、3′-甲氧基葛根素和牛蒡苷的含量。方法采用Agilent Eclipse XDB-C 18色谱柱(4.6 mm×250 mm,5μm),0.05%磷酸-乙腈梯度洗脱,柱温40℃。结果绿原酸、葛根素、3′-甲氧基葛根素和牛蒡苷4个组分的进样质量范围分别在0.0039~0.2353μg(r=0.9999)、0.0651~3.9066μg(r=0.9997)、0.0190~1.1404μg(r=0.9998)、0.0522~3.1331μg(r=0.9998)与峰面积呈良好的线性关系;分离度良好;加样回收率分别为99.3%、98.6%,99.4%,99.9%,RSD分别为0.7%,1.3%,1.8%,0.9%。结论方法学验证结果表明本法准确,灵敏度高,重复性好,能为鸢都感冒颗粒全面的质量控制提供参考。
Objective To develop a new method for simultaneous determination of chlorogenic acid,puerarin,3′-methoxypuerarin and arctiin in Yuandu Ganmao Granules by HPLC-DAD.Methods The chromatographic separation were performed on an Agilent Eclipse XDB-C 18 column(4.6 mm×250 mm,5μm)with gradient elution by 0.05%phosphoric acid-acetonitrile.The column temperature was 40℃.Results The ranges for linear correlation of chlorogenic acid,puerarin,3′-methoxypuerarin and arctiin were 0.0039~0.2353μg(r=0.9999),0.0651~3.9066μg(r=0.9997),0.0190~1.1404μg(r=0.9998),0.0522~3.1331μg(r=0.9998),respectively.Excellent chromatographic separation was achieved.The average recoveries were 99.3%,98.6%,99.4%,99.9%,with RSD 0.7%,1.3%,1.8%,0.9%,respectively.Conclusion This method was accurate,sensitive,reproducible,which can be used for the overall assessment of the quality of Yuandu Ganmao Granules.
