摘要
目的建立一种高尿酸血症联合2型糖尿病的大鼠模型,并从多方面探讨高尿酸血症合并2型糖尿病发病过程中各项指标的关联性。方法取96只SD大鼠随机分为A组(空白对照)、B组(每日脂肪乳1 mL/kg灌胃)、C组(每日腹腔注射200 mg/kg氧嗪酸钾)、D组(每日250 mg/kg腺嘌呤+150 mg/kg乙胺丁醇灌胃)、E组(每日腹腔注射200 mg/kg氧嗪酸钾并脂肪乳1 mL/kg灌胃)、F组(每日250 mg/kg腺嘌呤+150 mg/kg乙胺丁醇灌胃,间隔8 h后脂肪乳1 mL/kg灌胃),B组及E、F组于干预第14日及第15日分别按照120、100 mg/kg腹腔注射四氧嘧啶,于二次注射后第2日晚各组实验动物均禁食12 h,进行糖耐量试验。于第3日晚禁食12 h后,麻醉取血检测血尿酸(UA)、肌酐(Scr)、尿素氮(BUN)、血糖(GLU)、甘油三酯(TG)、胆固醇(TC),处死动物,取下肾脏、胰腺,制作相关脏器病理切片并进行比较。结果E组大鼠GLU、UA、BUN、TG、TC水平较A组明显上升(P<0.05),F组GLU、UA、BUN、Scr、TC、TG较A组明显上升(P<0.05)。E、F组大鼠胰腺均出现了病理损伤,F组大鼠还出现了肾脏病理损伤。结论E、F组高尿酸血症合并2型糖尿病大鼠的各项指标均符合2种疾病的临床表现,2种方法均可用于该模型的建立。
Objective To establish a rat model of hyperuricemia combined with type 2 diabetes mellitus and to explore the correlation of various indexes in the pathogenetic process of hyperuricemia combined with type 2 diabetes mellitus.Methods 96 SD rats were randomly divided into group A(control group),group B(daily 1 mL/kg fat emulsion was given by gavage),group C(daily intraperitoneal injection of oteracil potassium 200 mg/kg),group D(250 mg/kg daily adenine+150 mg/kg ethambutol were given by gavage),group E(200 mg/kg daily intraperitoneal injection of oteracil potassium and 1 mL/kg fat emulsion was given by gavage)and group F(250 mg/kg daily adenine+150 mg/kg ethambutol were given by gavage,after 8 hours,1 mL/kg fat emulsion was given by gavage).The experimental animals in group B,combined with group E and F were intraperitoneally injected with alloxan at 120 mg/kg and 100 mg/kg respectively on the 14th and 15th day of intervention,and fasted for 12 h on the 2nd night after the second injection to conduct glucose tolerance test.After fasting for 12 h on the evening of the third day,blood samples were taken under anesthesia to detect UA,BUN,Scr,GLU,TG and TC.Afterthe animals were killed,the kidney and pancreas were collected to make pathological section.Results The levels of GLU,UA,BUN,TG and TC in rats in group E were significantly higher than those in group A(P<0.05),and GLU,UA,BUN,Scr,TC and TG in group F were significantly higher than those in group A(P<0.05).Rats in group E and F showed pathological damage to the pancreas,and rats in group F also showed pathological damage to the kidney.Conclusion The indexes of hyperuricemia combined with type 2 diabetes mellitus in the two groups are consistent with the clinical manifestations of the two diseases,indicating that both methods can be used for the establishment of the model.
