摘要
目的基于网络药理学方法对马齿苋治疗细菌性痢疾的活性成分及作用机制进行分析,探讨多成分-多靶点-多通路的网络关系。方法从TCMSP、TCMID、BATMAN-TCM数据库获取马齿苋的成分,并通过TCMSP数据库的药物动力学参数筛选其活性成分及获取其作用靶点的信息。通过CTD、TTD、DiSGeNET和DrugBank这4大数据库获取细菌性痢疾相关靶点,并通过String数据库筛选其细菌性痢疾特异性主要靶点。将马齿苋靶点与细菌性痢疾靶点进行映射,得到马齿苋作用于细菌性痢疾的预测靶点。预测的共有靶点利用Cytoscape3.7.1软件构建网络,并通过DAVID数据进行GO基因功能和KEGG信号通路分析。结果马齿苋的山奈酚、槲皮素、木犀草素、β-胡萝卜素和β-谷甾醇等成分主要通过肿瘤坏死因子(TNF)信号通路作用于TNF、环加氧酶2(PTGS2)和白细胞介素-1β(IL-1β)等潜在靶点而发挥治疗细菌性痢疾的作用。结论马齿苋可能通过减轻肠黏膜损伤和抑制肠道炎性反应而发挥治疗细菌性痢疾的作用。
Objective To analyze the active components and mechanism of Portulaca oleracea L.in the treatment of bacillary dysentery based on the method of network pharmacology,and to explore the network relationship of multi-component,multi-target and multi-channel.Methods The components of Portulaca oleracea L.were obtained from TCMSP,TCMID and BATMAN-TCM database,and then the active components and target information were screened by the pharmacokinetic parameters of TCMSP database.The related targets of bacillary dysentery were obtained by CTD,TTD,DiSGeNET and Drugbank,and the main targets of bacillary dysentery were screened by string database.By mapping Portulaca oleracea L.target with bacillary dysentery target,the prediction target of Portulaca oleracea L.acting on bacillary dysentery was obtained.The predicted common target was constructed by using the software of Cytoscape3.7.1,and the function of GO gene and the signal pathway of KEGG were analyzed by the data of DAVID.Results The potential components of Portulaca oleracea L.,such as Kaempferol,Quercetin,Luteolin,β-carotene andβ-sitosterol,played a role in the treatment of bacillary dysentery mainly through tumor necrosis factor(TNF)signaling pathway acting on potential targets such as TNF,prostaglandin endoperoxide synthase 2(PTGS2)and interleukin-1β(IL-1β).Conclusion Portulaca oleracea L.may play a role in the treatment of bacillary dysentery by reducing intestinal mucosa damage and inhibiting intestinal inflammation.
