解表宽中化浊行血法对慢性阻塞性肺疾病大鼠气道重塑的影响研究 被引量:4

Study on the effect of Jiebiao Kuanzhong Huazhuo Xingxue method on airway remodeling in rats with chronic obstructive pulmonary disease

摘要 目的观察康肺散结化浊行血汤对慢性阻塞性肺疾病(COPD)大鼠血清基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)表达及肺组织病理学的影响,探讨解表宽中、化浊行血法对COPD大鼠气道重塑的治疗机制。方法选择40只清洁级雄性Wistar大鼠,按随机数字表法分为正常组、模型组和康肺散结化浊行血汤高剂量组、康肺散结化浊行血汤低剂量组,每组10只。除正常组外,其余组采用烟熏加脂多糖气道滴注法制作COPD大鼠模型。造模成功后,高、低剂量康肺散结化浊行血汤组分别给予康肺散结化浊行血汤10 mL/kg、20 mL/kg灌胃,正常组和模型组给予生理盐水10 mL/kg灌胃。灌胃30 d后,ELISA法检测各组大鼠血清MMP-9和TIMP-1水平,电镜法观察各组大鼠肺切片组织形态。结果与正常组比较,模型组大鼠血清MMP-9、TIMP-1水平及MMP-9/TIMP-1显著升高(P均<0.05);与模型组比较,高、低剂量康肺散结化浊行血汤组大鼠血清MMP-9、TIMP-1水平及MMP-9/TIMP-1均显著降低(P均<0.05),肺组织切片所示肺气肿明显减轻,且与剂量正相关。结论解表宽中、化浊行血法可降低COPD模型大鼠血清MMP-9、TIMP-1水平,减轻肺组织气道破坏,改善气道重塑,对COPD有一定治疗作用。 Objective It is to observe the effect of Kangfei Sanjie Huazhuo Xingxue decoction on the expression of matrix metalloproteinase-9(MMP-9),matrix metalloproteinase inhibitor-1(TIMP-1)in serum and lung histopathology of chronic obstructive pulmonary disease(COPD)rats,and to discuss the therapeutic mechanism of Jiebiao Kuanzhong Huazhuo Xingxue method on airway remodeling in COPD rats.Methods Forty male Wistar rats of clean grade were randomly divided into normal control group,COPD model group and high and low dose of Kangfei Sanjie Huazhuo Xingxue decoction group with 10 rats in each group.COPD rat model was established by fumigation and lipopolysaccharide airway drip in every group except for the normal group.After the successful establishment of the model,the high-dose and low-dose Chinese medicine groups were given Kangfei Sanjie Huazhuo Xingxue decoction 10 and 20 mL/kg daily respectively by gavage,while the control group and the model group were given 10 mL/kg normal saline by gavage.After intragastric administration for 30 days,the levels of serum MMP-9 and TIMP-1 of rats in each group were detected by ELISA,and the morphology of lung sections of rats in each group was observed by electron microscope.Results Compared with the normal control group,the levels of serum MMP-9,TIMP-1 and MMP-9/TIMP-1 in the Chinese medicine model group were significantly increased(P<0.05);compared with the model group,the levels of serum MMP-9,TIMP-1 and MMP-9/TIMP-1 in high and low dose of Kangfei Sanjie Huazhuo Xingxue decoction groups were significantly decreased(P<0.05),and emphysema in lung tissue sections was significantly reduced,and the results was positively correlated with the dose.Conclusion Jiebiao Kuanzhong Huazhuo Xingxue method can reduce serum MMP-9 and TIMP-1,alleviate lung tissue airway damage and improve airway remodeling in COPD model rats.It has certain therapeutic effect on COPD.
