中国蜂胶及其主要组分对两种肺癌细胞作用及机制研究 被引量:3

Study on the antitumor activities and mechanisms of Chinese propolis and its major constituents by two lung cancer cells

摘要 蜂胶具有较好的抗肿瘤功效,但其功效成分及其作用机理还未完全阐明。本文研究了中国蜂胶乙醇提取物(EECP)(100μg/ml)及其主要组分(咖啡酸苯乙酯、白杨素、芹菜素、高良姜素)(80μM)抑制两种肺癌细胞(A549和H1299)的作用功效及机理。通过transwell实验,发现EECP及其主要组分显著抑制A549和H1299细胞的浸润;通过Hoechst33258染色和Western blotting检测,发现EECP及其主要组分显著促进两种肺癌细胞的凋亡,上调caspase 3水平,降低未剪切PARP蛋白水平,而EECP及其主要组分促进A549和H1299两种肺癌细胞凋亡与上调肿瘤细胞内活性氧水平、降低线粒体膜电位有关。蜂胶及其主要组分是潜在的抗肿瘤天然活性物质。 Propolis has good antitumor activity,but its biological constituents and mechanism of propolis inhibiting tumor cells proliferation has not been fully elucidated.The antitumor activities and mechanisms of Chinese propolis ethanol extract(EECP)(100μg/ml)and its main constituents(caffeic acid phenethyl ester,chrysin,apigenin,and galangin)(80μM)on two different lung cancer cells(A549 and H1299)were studied in the present article.EECP and its major constituents obviously inhibited two lung cancer cells invasion by transwell study.Furthermore,they also significantly promoted apoptosis of two lung cancer cells by Hoechst33258 staining and western blotting test.EECP and its major constituents apparently up-regulated the caspase 3 levels and down-regulated the levels of uncleaved PARP.The proapoptotic effects of EECP and its major constituents were probably related with increasing reactive oxygen species and decreasing mitochondria membrane potential.In all,propolis and its major constituents are potential antitumor natural products.
