云南松针精油的提取及抗氧化活性研究 被引量:16

Optimization of extraction process and antioxidant activity of essential oil from needles of Pinus yunnanensis

摘要 以云南松针为原料,采用共水法提取松针精油,通过单因素正交试验优化提取工艺,测定精油的理化性质和挥发性成分,并考察松针精油的抗氧化活性。结果表明,共水法提取云南松针精油的最优工艺为NaCl用量为8%,料液比为1∶7g/mL,提取时间为4h,浸泡时间为2h,出油率为1.33%;松针精油中共鉴定出15种化合物,较水蒸气蒸馏法提取的挥发油成分单一,主要为单萜和倍半萜类化合物,其中α-蒎烯含量高达64.23%,且其折光度、相对密度、旋光度均符合国家标准要求;精油具有良好的抗氧化活性,对ABTS+·、DPPH·、OH·(结晶紫法)、OH·(水杨酸法)的半数清除率IC50值分别为4.027、47.294、2.492、0.078mg/mL,其中对OH·的清除能力与维生素C无显著差异。而松针精油对Fe3+的还原能力较维生素C的差,且存在显著差异。本文通过优化了共水法提取松针精油的工艺,并测定该方法获得的松针精油的理化性质和主要挥发性成分,考察其抗氧化活性,以期为云南松针精油在食品、医药、化工等领域的开发利用提供理论依据。 Needles of Pinus Yunnanensis was used as raw material,the essential oil of pine needles was extracted by co-distillation.The extraction process was optimized by single factor orthogonal test.Physicochemical properties and volatile components of essential oil were determined,and antioxidant activity of essential oil was investigated.The results showed the optimum process of extracting essential oil was:NaCl 8%,material-to-liquid ratio 1:7 g/mL,extraction time 4 h,soaking time 2 h.Under the above condition,the yield of essential oil was 1.33%.A total of 15 compounds were identified and were mainly composed of monoterpenoids and sesquiterpenes.The content ofα-pinene was as high as 64.23%,the refraction,relative density and rotation all meet national standards.The essential oils exhibited excellent antioxidant activity,the IC50 values to ABTS+·,DPPH·,OH·(crystal violet),and OH·(salicylic acid)were 4.027,47.294,2.492,0.078 mg/mL,respectively.The removal capacity for OH·was not significantly different from that of Vc,while the reducing capacity of the essential oil to Fe3+was worse than that of Vc,with a significant difference.In summary,the process of extracting essential oil from needles of Pinus Yunnanensis by co-distillation method was optimized,the physicochemical properties and main volatile components of the essential oil were determined,and its antioxidant activity was investigated.The study provides a theoretical basis for the development and utilization of pine needle essential oil in food,medicine,chemical industry.
