摘要
目的:针对刺玫果汤剂,基于HPLC图中色谱峰信息,利用化学计量学方法分类刺玫果药材,以抗氧化作用为指标对分类出的刺玫果药材进行优选,最终建立刺玫果指纹图谱和函数质量表达式。方法:采用HPLC法对成分进行峰面积采集,运用聚类分析法和主成分分析法分类样品;以抗氧化作用为指标,对刺玫果分类出的样品进行评价,确定最优刺玫果药材,最终建立刺玫果汤剂指纹图谱,同时利用判别分析建立质量数学表达式。结果:不同采集地样品分为3类,其中I类(15批)刺玫果样品,抗氧化作用最高,以1,1-二苯基-2-三硝基苯肼法测定得到的半数抑制率(IC50)对应的质量浓度为0.244 mg·mL-1,以2,2’-联氮-双-3-乙基苯并噻唑啉-6-磺酸法测定得到的IC50对应的质量浓度为0.204 mg·mL-1,以抗氧化作用为标准获得质量可靠、稳定的刺玫果样品,最终建立了指纹图谱,同时得到函数质量表达式Y=-35.846-0.014X1-0.10X2+0.042X3-0.023X4+0.102X5+0.014X6-0.050X7+0.066X8-0.106X9-0.038Z10+0.010X11+0.09X12-0.040X13+0.071X14+0.051X15-0.044X16,函数重心为(-20.550,0.984)。结论:本研究探讨了对药材质量评价的方法,该方法科学,更具有实用意义。
Objective:To establish the fingerprint and mathematical quality expression by using the chemometrics method to classify Dahurian rose fruit and taking the antioxidant effect as the index to optimize the classified Dahurian rose fruit basing on the chromatographic peak information in the HPLC figure. Methods:HPLC was used to collect the peak area of 16 components,and cluster analysis and principal component analysis were used to classify samples. The antioxidant effect was used as the index to evaluate the classified samples of Cimeiguo and chose the best material. Finally,the fingerprint of Cimeiquo decoction was established and the mathematical expression of quality was constructed by discriminant analysis. Results:Samples from different places were divided into three categories. Cimeiquo samples of class Ⅰ(15 batches)claimed the highest antioxidant effect. The corresponding concentrations of IC50 determined by 1,1 diphenyl-2-picrylhydrazyl and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid methods were 0.244 mg·mL-1 and 0.204 mg·mL-1,respectively. Dahurian rose fruit samples on the basis of antioxidant effect quality was reliable and stable. In the end a fingerprint was established. The functional expression for quality was Y=-35.846-0.014 X1-0.10 X2+0.042 X3-0.023 X4+0.102 X5+0.014 X6-0.050 X7+0.066 X8-0.106 X9-0.038 X10+0.010 X11+0.09 X12-0.040 X13+0.071 X14+0.051 X15-0.044 X16. Function center of gravity range was(-20.550,0.984). Conclusion:This study discussed the method of evaluating the quality of medicinal materials.
