摘要
目的:探讨寿胎丸对复发性流产模型大鼠蜕膜细胞水通道蛋白(AQP)1、2表达的影响。方法:取复发性流产模型大鼠胚胎着床部位的蜕膜组织进行原代和传代培养,分别用寿胎丸含药血清、雌激素(E2)及孕激素(P)进行干预,另设空白对照组,4组药物对培养的蜕膜细胞作用72h后,通过qRT-PCR及Western blot方法检测蜕膜细胞中AQP1、AQP2 mRNA及蛋白的表达。结果:与空白对照组比较,寿胎丸组及P组AQP1、AQP2 mRNA及蛋白的表达均显著提高(P<0.05),E2组AQP1、AQP2 mRNA及AQP2蛋白的表达显著提高(P<0.01)。P比寿胎丸更显著上调AQP1 mRNA及蛋白的表达(P<0.01)。E2、P均较寿胎丸显著上调AQP2 mRNA的表达(P<0.01)。结论:寿胎丸可能通过上调复发性流产大鼠蜕膜组织AQP1、AQP2 mRNA及蛋白的表达水平,改善妊娠结局。寿胎丸对AQP蛋白的调节,说明“肾主水”与“肾主生殖”之间存在相关性。
Objective:To investigate the effects of Shoutai Pills on the expression of aquaporin 1(AQP1),aquaporin 2(AQP2)in decidual tissue of maternal-fetal interface in rats with recurrent abortion.Methods:The decidual tissue of the embryonic implantation site of rats with kidney deficiency and luteal insufficiency was cultured in primary and subculture.Estrogen(E2),progesterone(P)and the serum containing Shoutai Pills were used to intervene.A blank control group was set up.qRT-PCR and Western blot were used to detect the expression of AQP1,AQP2 mRNA and protein in decidual cells after 72h of action of four groups of drugs on cultured decidual cells.Results:Compared with the control group,the expression of AQP1,AQP2 mRNA and protein was significantly increased in Shoutai Pills group and P group(P<0.05);E2 group significantly increased the expression of AQP1,AQP2 mRNA and AQP2 protein(P<0.01).The expression of AQP1 mRNA and protein in P group was significantly higher than that in Shoutai Pills group(P<0.01).The expression of AQP2 mRNA in E2 and P groups was significantly higher than that in Shoutai Pills group(P<0.01).Conclusion:Shoutai Pills may improve pregnancy outcomes by up-regulating the expression of AQP1,AQP2 proteins in decidua in rats with recurrent abortion.The regulation of AQP by Shoutai Pills indicates that there is a correlation between‘kidney dominates water’and‘kidney dominates reproduction’.
