摘要
真核翻译起始因子eIF5A与植物的生长发育及抗逆生长密切相关。以玉米弯孢菌抗性自交系Mo17和感性自交系黄早四为试验材料,克隆得到eIF5A基因,并对其进行生物信息学分析。结果表明:在Mo17和黄早四中分离的eIF5A基因均包含480bp的ORF,编码160个氨基酸,分子量分别为17.50和17.48KD,等电点均为5.61。通过对氨基酸序列的分析发现,eIF5A基因编码的蛋白质为非跨膜稳定的亲水性蛋白,存在15个磷酸化位点,与高粱的相似度最高,并且该蛋白的保守结构域属PLN103107超级家族。
Eukaryotic translation initiation factor(eIF5 A)is closely related to plant growth,development and stress tolerance.The resistant inbred line Mo17 and the susceptible inbred line Huangzao4 to Curvularia lunate(Walker)Boedijn as materials,cloning and bioinformatic analysis of eIF5 A genes were conducted.The results showed that the eIF5 A genes isolated from Mo17 and Huangzao4 both involved ORFs comprising of 480 base pairs,encoding aprotein comprising of 160 amino acids with molecular weights of 17.50 KD and 17.48 KD,respectively,and isoelectric points of 5.61.The results of amino acid sequence analysis revealed that eIF5 Awas a nontransmembrane stable and hydrophilic protein with 15 phosphorylation sites,its similarity to eIF5 A protein from sorghum was the highest,and its conserved domain belonged to the PLN103107 super family.
