摘要
目的研究小鼠脑干和小脑血管的形态学特点,为构建脑血管疾病动物模型提供形态学依据。方法1%戊巴比妥钠腹腔内注射麻醉小鼠,开胸行左心室插管术,依次用0.9%氯化钠溶液、4%多聚甲醛、墨汁灌流,取脑,冰冻切片和尼氏染色,数字切片扫描观察小鼠脑干和小脑血管的形态学特点。结果脑干和小脑血管面积所占比例:延髓(8.03±1.37)%、脑桥(8.75±0.99)%、中脑(11.87±1.45)%、小脑(12.24±1.90)%,两两比较差异无统计学意义(P>0.05);神经核团内神经元胞体聚集区血管数量多、密度大,密集分布于神经元胞体间;小脑蚓神经元细胞数量多,排列紧密。结论墨汁灌注结合数字切片扫描能清晰显示小鼠脑干和小脑血管形态学特点,能为脑血管疾病的动物模型研究提供形态学参考。
Objective To study the morphological characteristics of brain stem and small cerebrovascular in mice,in order to provide the morphological basis for animal models of cerebrovascular diseases.Methods Mice were anesthetized by intraperitoneal injection of 1%sodium pentobarbital.Left ventricular intubation was performed after thoracotomy and the mice were perfused with 0.9%sodium chloride,4%paraformaldehyde,and ink in sequence.Brain was taken,and after frozen section and nissl staining,the morphological features of brain stem and small cerebral vessels were observed by digital section scanning.Results The proportion of brainstem and small cerebrovascular area(area%):medulla oblongata(8.03±1.37),pons(8.75±0.99),midbrain(11.87±1.45),cerebellum(12.24±1.90).Pairwise comparison showed no statistical significance(P>0.05).The number and density of blood vessels in the aggregation area of neurons in the nucleus are large,and they are densely distributed among neurons.Cerebellar worm neurons are numerous and closely packed.Conclusion The morphological features of mouse brain stem and cerebellum can be clearly shown by the combination of ink perfusion and digital section scanning which can provide a morphological reference for the study of animal model of cerebrovascular diseases.
