摘要
目的探索不同剂量匹罗卡品单次腹腔注射对诱发小鼠癫痫及对海马损伤的影响,为癫痫建模提供参考。方法将100只健康的C57BL/6小鼠随机分为正常对照组、3个不同剂量匹罗卡品(250 mg/kg、290 mg/kg、350 mg/kg)腹腔注射组。比较3组的建模成功率、死亡率,通过免疫组化染色观察神经元和胶质细胞的数量变化,分析海马区组织的病变情况。结果匹罗卡品250 mg/kg组小鼠的Racine评分4级以上发作率为12%,无死亡;290 mg/kg组小鼠的4级以上发作率为70%,死亡率为32%;350 mg/kg组小鼠的4级以上发作率为86%,死亡率为56%。免疫组化染色结果显示:与正常对照组相比,3个不同剂量匹罗卡品组小鼠诱发癫痫后,神经元数量均显著减少,小胶质细胞和星形胶质细胞数量均显著增加;但250 mg/kg组小鼠的活化小胶质细胞数量无显著变化,而其他两组显著增多。结论匹罗卡品290 mg/kg单次注射诱发小鼠癫痫模型的成功率较高、死亡率较低,且海马区损伤明显,可以作为癫痫小鼠建模的参考方法。
Objective To explore the effects of single injection of varying doses of pilocarpine on hippocampal injury in mice and to providea reference for creating the model of epilepsy.Methods A total of 100 healthy C57BL/6 mice were divided into the normal group and three treated groups with various doses of pilocarpine injection,namely 250 mg/kg,290 mg/kg and 350 mg/kg.After that,the success rate and mortality were compared.The lesions of hippocampus were detected by immunohistochemical staining for the neurons and glial cells.The pathological changes of hippocampus were analyzed.Results In this series,12%of mice could reach level 4(Racine scale)in 250 mg/kg group,and no mortality was found.In 290 mg/kg group,70%of mice could get to the level 4,and the mortality was 32%.In the 350 mg/kg group,86%of mice could reach level 4,and the mortality rate was 56%.Immunohistochemical staining results showed that the number of neurons in the hippocampus was significantly reduced compared with that in the normal group after injection of different concentrations of pilocarpine.Meanwhile,the number of microglia and astrocytes in the three groups was significantly higher than that in the normal group.However,there was no significant change in the number of activated microglia between the 250 mg/kg group and normal group,which was significantly increased in the other two groups.Conclusion The single injection of 290 mg/kg pilocarpine induced epilepsy model with high success rate,low mortality and obvious damage in hippocampus can be used as a reference method for creating the model of epilepsy.
