摘要
目的:研究半胱氨酸天冬氨酸蛋白酶(Caspase3)在红藻氨酸癫痫模型大鼠海马凋亡神经元中,对其已知底物真核生物起始因子2(eIF2α)的作用。方法:制备红藻氨酸诱导大鼠癫痫模型,记录模型发作行为及脑电图表现,以TUNEL法检测模型癫痫发作终止后12、24、48、72和96h海马神经元凋亡情况,同时以Westernblot法检测模型鼠海马eIF2α的表达变化及其与细胞凋亡之间的相关性。结果:在癫痫发作后12h出现凋亡神经元,随时间延长,海马神经元凋亡数不断增加,72h达高峰;发作后24h海马神经元出现了Caspase3酶切eIF2α产生的片段,并逐渐变浓,至72h。结论:癫痫模型大鼠海马中Caspase3酶切eIF2α与神经元凋亡的发生时间一致,提示在癫痫模型大鼠海马神经元凋亡中eIF2α被Caspase3酶切。
Objective:This investigation is aimed to explore the possible role of Caspase-3 on eIF2α in the hippocampal apoptotic neuron of Kainic acid(KA)-induced epileptic rat. Methods:The KA epileptic model was established and the apoptotic neurons in the hippocampus of the model rats was detected by TUNEL method. Western blot is used to detect the change of eIF2α in hippocampal neurons of epileptic model rats. Results:The apoptotic rate reached a peak in 72 hours after epileptic discharge. The cleavage of eIF2α by Caspase-3 was detected in hippocampal neurons of epileptic model. Conclusion:Recurrently epileptic discharge leads neurons to apoptosis.The cleavage of eIF2α by Caspase-3 accompanies the apoptosis.
