摘要
目的:采用酶法优选余甘子多酚类化合物的提取工艺。方法:在单因素试验的基础上,采用正交设计进行提取条件的优化,以总多酚的提取率为考察指标,采用福林-酚比色法进行总多酚的含量测定,确定了提取工艺的最佳参数。结果:确定余甘子中总多酚的最佳提取工艺为:温度60℃,料液比为1∶30,纤维素酶和果胶酶的比例为1∶1,总加酶量为2%,pH值为4.0,恒温振荡(低速r=60~100/min),提取时间4 h,总多酚提取率为13.68%。结论:酶法提取优于传统溶剂提取法,操作简单可行,为余甘子总多酚的提取工艺提供了参考。
Objective To optimize the extraction process of polyphenols from Fructus Phyllanthus by enzymatic method.Methods Based on the results of single factor test,we designed the orthogonal experiment to optimize parameters.The content of total polyphenols was used as the investigation index and determined by Folin phenol colorimetric method,then obtain the optimum parameters of the extraction process.Results The optimum extraction process of total polyphenols was as follows:the temperature at 60℃,the ratio of liquid to material in 30∶1,the proportion of cellulase with pectinase about 1∶1,with the amount of adding enzyme 2%,and pH at 4.0,finally,extracting at constant temperature and constant shaking conditions(r=60~100/min)for 4 h,so the total polyphenol yield was 13.68%.Conclusion The enzymatic extraction was better than the traditional solvent extraction method,besides the operation was simple and feasible,and the experimental result provide a reference for the extraction process of total polyphenols from Phyllanthus.
