酶解法辅助提取钩藤中钩藤碱的工艺优选 被引量:8

Optimization of Extraction Process of Rhynchophylline from Uncariae Ramulus Cum Uncis by Enzymolysis Method

摘要 目的:优选酶解法辅助提取钩藤中钩藤碱的工艺条件,为该药材的资源利用提供参考。方法:采用HPLC测定钩藤碱含量,流动相甲醇-水(60∶40),检测波长254 nm。以钩藤碱提取率为指标,在单因素试验基础上,通过正交试验考察加酶量、酶解温度、酶解时间、酶解p H对酶解提取工艺的影响。结果:最佳酶解提取工艺条件为纤维素酶用量1%,p H 4.5,酶解温度50℃,酶解时间2 h。钩藤碱提取率0.54%,较加热回流法提高了54.29%。结论:酶解法有利于提高钩藤碱的得率,优选的提取工艺稳定可行,适合推广于药材中生物碱类成分的提取。 Objective: To optimize extracting process of rhynchophylline from Uncariae Ramulus Cum Uncis by enzymolysis method. Method: HPLC was adopted to determine the content of rhynchophylline with mobile phase of methanol-water(60 ∶ 40) and detection wavelength at 254 nm. With extraction rate of rhynchophylline as index, on the basis of single factor tests, effects of enzyme dosage, p H, enzymolysis temperature and time on extraction process were investigated by orthogonal test. Result: Optimal extraction conditions were as follows: dosage of cellulase 1%,p H of 4. 5,enzymolysis temperature of 50 ℃,enzymolysis time of 2 h. Extraction rate of rhynchophylline was 0. 54%,it increased 54. 29% by comparing with heating reflux method. Conclusion: Enzymolysis method is suitable for extracting rhynchophylline from Uncariae Ramulus Cum Uncis,this optimized extraction process is stable and feasible,it is worthy of popularization and application for extraction of alkaloids from herbs.
