Study on the Quality Standard for Jincao Xiaodu Granules

摘要 目的:建立金草消毒颗粒的质量标准。方法:采用TLC法鉴别金草消毒颗粒中3味中药饮片:肿节风、白花蛇舌草、半枝莲;同时以HPLC法测定异嗪皮啶、迷迭香酸含量,色谱柱:Aglient Zorbax SB-C18柱(250 mm×4.6 mm,5μm),流动相:乙腈-0.2%磷酸(25∶75)等度洗脱,流速:1.0 ml·min^-1,检测波长:342 nm,柱温:25℃。结果:薄层色谱斑点清晰,分离度好,阴性无干扰;异嗪皮啶和迷迭香酸分别在5.23~31.39μg·ml^-1(r=0.9984)、2.62~30.10μg·ml^-1(r=0.9997)范围内线性关系良好,平均回收率分别为89.18%(RSD=1.51%)、107.68%(RSD=0.85%)(n=6)。结论:本研究建立的质量控制方法快速、简单、专属性强,适用于金草消毒颗粒的质量控制。 Objective:To establish the quality standard for Jincao Xiaodu granules.Methods:TLC was used for identifying Sarcandra glabra,Hedyotis diffusa Willd and Scutellaria barbata in Jincao Xiaodu granules.The contents of isoxazinidine and rosemic acid were analyzed by high performance liquid chromatography on an Aglient Zorbax SB-C18 column(250 mm×4.6 mm,5μm)with the mobile phase of acetonitrile-0.2%phosphoric acid(25∶75)with isocratic elution at a flow rate of 1.0 ml·min^-1.The detection wavelength was 342 nm and the column temperature was controlled at 25℃.Results:The TLC spots were clear and well-separated that could be distinguished from the negative samples.Isoxazinidine and rosemic acid was linear respectively within the range of 5.23-31.39μg·ml^-1(r=0.9984)and 2.62-30.10μg·ml^-1(r=0.9997),and the average recovery was 89.18%(RSD=1.51%)and 107.68%(RSD=0.85%)(n=6),respectively.Conclusion:The method established in the study is fast,simple and specialized,which can be used for the quality control of Jincao Xiaodu granules.
