摘要
目的:通过建立免疫性血小板减少性紫癜(ITP)小鼠动物模型,观察正源方对模型小鼠血小板系统及免疫功能的影响,探讨正源方的免疫调节机制。方法:60只BALB/c小鼠随机分为正常对照组、模型对照组、复方皂矾丸组、正源方低、中、高剂量组。除正常对照组外,其余各组小鼠采用隔日腹腔注射豚鼠抗小鼠血小板血清(GP-APS)建立小鼠ITP模型。于造模第8天开始,复方皂矾丸组按1.68g/kg灌胃,正源方低、中、高剂量组按3、6和12g/kg灌胃,正常对照组、模型对照组灌服生理盐水,连续给药7天,观察各组小鼠外周血象的变化。实验结束后,计算脾脏指数,进行骨髓涂片,显微镜下观察巨核细胞计数和分类。结果:与正常对照组比较,通过腹腔注射GP-APS,模型组小鼠外周血小板计数显著降低,脾脏指数明显升高,骨髓巨核细胞总数、颗粒巨核细胞比例增高,产板巨核细胞比例明显降低;与模型对照组比较,正源方各剂量组(3g/kg,6g/kg,12g/kg)小鼠的血小板计数明显增高,3g/kg,6g/kg脾脏指数显著下降、巨核细胞数减少、产板巨核细胞比例增高,3g/kg颗粒巨核细胞比例显著下降;骨髓涂片显示正源方对巨核细胞的成熟具有一定的恢复作用。结论:正源方能够明显促进ITP模型小鼠外周血小板数和脾脏的恢复,促使骨髓巨核细胞成熟。
Objective: To observe the effect of Zhengyuan Prescription on platelet system and immune function in mice with immune thrombocytopenic purpura( ITP) and discuss its immunoregulatory mechanism. Methods: 60 BALB /c mice were randomly divided into control group,model group,Zaofan Pills group,and three Zhengyuan Prescription groups with low,medium,and high dose,respectively. Except the control group,the other mice were injected intraperitoneally guinea pig-antimouse platelet serum( GP-APS) every other day. 7 days later,the mice of Zaofan Pills group were given Zaofan Pills( 1. 68g /kg),the mice of the three Zhengyuan Prescription groups were given Zhengyuan Prescription for3,6,12g /kg respectively for seven days,while the remaining groups were given physiological saline with equal volumes. The peripheral blood of all the groups was observed every day. After the experiment,all the spleens were weighed and the organ index was measured. The amounts and morphology of megakaryocyte were observed under the microscope. Results: By intraperitoneal injection of GP-APS,compared with the control group,the peripheral blood platelet counts of model group continually decreased,spleen index,the bone marrow megakaryocyte counts and granular megakaryocyte ratio significantly increased,and platelet-producing megakaryocyte ratio reduced obviously. Compared with the model group,the number of peripheral platelet significantly increased at the doses of 3 ~ 1 2g /kg of Zhengyuan Prescription. Spleen index,megakaryocyte counts and granular megakaryocyte ratio significantly decreased with Zhengyuan Prescription at 3g /kg and 6g /kg. The plateletproducing megakaryocyte ratio increased obviously by Zhengyuan Prescription at 3g /kg. The results of bone marrow smear showed that Zhengyuan Prescription had a superior therapeutic effect on the mature of megakaryocytic cells. Conclusion: Zhengyuan Prescription was able to improve platelet counts,the recovery of peripheral blood platelet and spleen and the mature of megakaryocyte of mice with immune thrombocytopenic purpura. Above all,the treatment effect of Zhengyuan Prescription was proved in the ITP mice.
