摘要
目的:建立HPLC梯度洗脱法同时测定小儿胃宝片中尿囊素、山药素Ⅰ、原花青素B2、原花青素C1、牡荆素葡萄糖苷、牡荆素鼠李糖苷和牡荆素的含量。方法:采用Agilent Zorbax SB C18色谱柱(250 mm×4.6 mm,5μm),柱温35℃,流动相A为甲醇-乙腈(1∶2),流动相B为0.05%甲酸水溶液,梯度洗脱,流速1.0 ml·min^-1,检测波长为224 nm(检测尿囊素、山药素Ⅰ)、280 nm(检测原花青素B2、原花青素C1)和350 nm(检测牡荆素葡萄糖苷、牡荆素鼠李糖苷、牡荆素)。结果:尿囊素、山药素Ⅰ、原花青素B2、原花青素C1、牡荆素葡萄糖苷、牡荆素鼠李糖苷、牡荆素分别在9.87~197.40μg·ml^-1,1.59~31.80μg·ml^-1,2.46~49.20μg·ml^-1,1.38~27.60μg·ml-1,1.96~39.20μg·ml^-1,14.47~289.40μg·ml^-1,0.79~15.80μg·ml^-1范围内线性关系良好(r≥0.9992),平均加样回收率分别为99.35%,97.84%,98.07%,98.53%,99.16%,100.21%,96.97%,RSD分别为0.94%,1.20%,1.61%,0.65%,0.88%,0.82%,1.35%(n=9)。结论:该方法操作便捷、重复性好,可作为小儿胃宝片质量控制的方法。
Objective:To establish an HPLC gradient elution method for determining the contents of allantoin,batatasin I,procyanidin B2,procyanidin C1,vitexin-4"-O-glucoside,rhamnosylvitexin and vitexin in Xiaoer Weibao tablets simultaneously.Methods:An Agilent Zorbax SB C18(250 mm×4.6 mm,5μm)chromatographic column was used,the mobile phase A was methanol-acetonitrile(1∶2)and the mobile phase B was 0.05%formic acid solution with gradient elution.The flow rate was set at 1.0 ml·min^-1 and the column temperature was set at 35℃.The detection wavelengths were 224 nm for allantoin and batatasin I,280 nm for procyanidin B2 and procyanidin C1,and 350 nm for vitexin-4"-O-glucoside,rhamnosylvitexin and vitexin.Results:Allantoin,batatasin I,procyanidin B2,procyanidin C1,vitexin-4"-O-glucoside,rhamnosylvitexin and vitexin showed good linear relationship within the range of 9.87-197.40μg·ml^-1,1.59-31.80μg·ml^-1,2.46-49.20μg·ml^-1,1.38-27.60μg·ml-1,1.96-39.20μg·ml^-1,14.47-289.40μg·ml^-1 and 0.79-15.80μg·ml^-1(r≥0.9992),and the average recovery was 99.35%,97.84%,98.07%,98.53%,99.16%,100.21%and 96.97%with the RSDs of 0.94%,1.20%,1.61%,0.65%,0.88%,0.82%and 1.35%(n=9),respectively.Conclusion:The method is easy to operate and repeatable,which can be used for the quality control of Xiaoer Weibao tablets.
