摘要
【目的】分析所收集洋紫荆(Bauhinia variegata)种质资源亲缘关系和遗传多样性。【方法】通过ISSR-PCR方法,对15个不同来源和特征的洋紫荆个体间亲缘关系和遗传多样性进行研究。【结果】ISSR实验共得到150个清晰位点,其中145位点具有多态性,总的多态性比率为96.67%,其中8条引物的扩增产物多态位点比率达到100%;洋紫荆材料的平均等位基因(Na)为1.9667±0.1801、平均有效等位基因数(Ne)为1.6847±0.2934、平枚Shannon信息指数(/)为0.5611±0.1639;亲缘关系分析表明,相同花色、相似开花量或相同来源地的不同个体间亲缘关系有时不显著。【结论】所收集洋紫荆资源形态特征具有丰富的遗传来源,亲缘关系复杂;同时,由于异地洋紫荆商品种苗活跃的流通导致不同来源种质资源亲缘关系复杂,种质资源高度杂合,育种过程亲本性状的纯化需较长周期。
[Objective]The affinity and genetic diversity of the collected Bauhinia variegata germplasm resources were analyzed.[Method]15 B.variegata individuals were systematically studied by ISSR-PCR method.[Result]A total of 150 clear loci were obtained in the ISSR experiment,of which 145 loci were polymorphic,and the total polymorphism ratio was 96.67%.Among them,the polymorphism ratios of amplification products of 8 primers reached 100%.The average allele(TVa)of Bauhinia materials was 1.9667±0.1801,the mean effective allele number(TVe)was 1.6847±0.2934,and the average Shannon information index(/)was 0.5611±0.1639.The phylogenetic analysis showed that there is not sometimes significant kinship among different individuals with the same flower color,similar flowering amount or the same source.[Conclusion]The morphological characteristics of the collected B.variegata resources had high hybridization,complex affinity,and at the same time,germplasm resources from different sources had a more complex affinity because of the active circulation of commodity seedlings,and the heterogeneity of germplasm resources was higher.As a result,the purification of parent traits in breeding process requires a long period.
