摘要
为探究舍内不同氨气浓度对鸭肝脏过氧化物酶体增殖物激活受体α(PPARα)、脂肪酸结合蛋白1(FABP1)和脂蛋白脂肪酶(LPL)基因的表达影响,将36只43周龄山麻鸭随机分为2组,即舍内氨气浓度10 mg·L^-1组和75 mg·L^-1组,饲养至10、20和30 d时采用荧光定量PCR法检测氨气对鸭肝脏PPARα及其信号通路中脂代谢相关基因FABP1和LPL表达的变化。结果表明:与10 mg·L^-1氨气组相比,75 mg·L^-1氨气组饲养至10、20、30 d时,鸭肝脏中PPARα、FABP1和LPL表达量均显著下降。相关性分析表明,在氨气应激状态下,PPARα表达量上升伴随LPL表达量显著上升,同时FABP1表达量上升,LPL表达量极显著上升。这说明高浓度氨气对鸭肝脏中PPARα、FABP1和LPL表达有显著影响,且PPARα和FABP1表达量与LPL表达量均呈正相关关系,这一研究有助于阐明鸭PPARα信号途径在氨气应激状态下对脂代谢调节的作用。
The effect of different ammonia concentration on PPARα,FABP1 and LPL gene expression in duck liver was investigated.In this study,thirty-six 43-week-old Shanma ducks were randomly divided into two groups,including the 10 mg·L^-1 group and 75 mg·L^-1 group.qRT-PCR was carried out to detect the PPARα,and the lipid metabolism related genes FABP1 and LPL in PPARαsignaling pathway in duck liver at 10 th,20 th and 30 th day during the trial.The results showed that the expressions of PPARα,FABP1 and LPL of ammonia exposure to 75 mg·L^-1 ammonia for 10,20 or 30 days in duck liver were respectively significantly lower than those of exposure to 10 mg·L^-1 ammonia for 10,20 or 30 days.Association analysis showed that the increase of PPARαexpression was accompanied by the significant increase of LPL expression under ammonia stress.Meanwhile,the increase of FABP1 expression was accompanied by the significant increase of LPL expression.These results indicated that high concentration of ammonia had a significant effect on the expression of PPARα,FABP1 and LPL gene in duck liver,and the expression of PPARαand FABP1 had a significant positive correlation with LPL.This study is helpful to elucidate the role of duck PPARαsignaling pathway in regulating the lipid metabolism under ammonia stress.
