摘要
建立了丙二醇甲醚醋酸酯(PMA)诱导的人急性单核白血病细胞(THP-1)炎症模型,通过细胞毒性试验确定乳铁蛋白对THP-1的安全质量浓度,考察了乳铁蛋白对THP-1分泌白介素6(IL-6)的影响。研究结果表明:乳铁蛋白在最大安全质量浓度为0.1 g/L时能有效抑制IL-6的分泌,使IL-6的相对分泌量降至21.7%,抑制率为78.3%,具有明显的抗炎活性。建立小鼠皮肤黑色素瘤细胞(B16-F10)模型,考察了乳铁蛋白对黑色素分泌的影响。研究结果表明:在最大安全质量浓度为1 g/L时,乳铁蛋白对黑色素的抑制率可达28.39%,具有明显的的美白活性。相同质量浓度条件下,0.5 g/L的乳铁蛋白和熊果苷对黑色素生成的抑制率分别为26.26%和12.15%,乳铁蛋白对黑色素表达的抑制高于功效对照原料熊果苷。因此乳铁蛋白具有明显的抗炎和美白活性,在化妆品领域有较好的应用前景。
The effects of lactoferrin on the secretion of interleukin-6(IL-6)in human acute monocytic leukemia cells(THP-1)induced by PMA were studied with an established inflammatory model.The results showed that,at the maximum safety mass concentration of 0.1 g/L,lactoferrin could effectively inhibit the secretion of IL-6 and reduce the relative level of release of IL-6 to 21.7%,and the inhibition rate was 78.3%,which showed obvious anti-inflammatory activity.The effects of lactoferrin on the secretion of melanin in mouse skin melanoma cells(B16-F10)were studied.The results showed that lactoferrin had significant whitening effect.At the same mass concentration,the effect of lactoferrin on reducing melanin content was better than that of arbutin:the inhibition rates of 0.5 g/L lactoferrin and arbutin on melanin production were 26.26%and 12.15%,respectively.The inhibition rate of lactoferrin on melanin production reached 28.39%at the highest safety mass concentration(1 g/L).Lactoferrin had demonstrable anti-inflammatory and whitening activity,and therefore it has a good application prospect in the field of cosmetics.
