二氢青蒿素对体外培养胃癌细胞株BGC-823周期蛋白D1 P16蛋白表达的影响 被引量:4

摘要 目的探讨二氢青蒿素(DHA)对人胃癌细胞BGC-823增殖的抑制作用及周期蛋白D1、P16蛋白表达的影响。方法以25~100μmol/L DHA干预人胃癌细胞株BGC-823细胞后通过四甲基偶氮唑盐(MTT)比色法测定细胞增殖率,吖啶橙-溴化乙定(AO-EB)染色观察细胞凋亡情况、流式细胞仪检测细胞凋亡周期、蛋白质印迹法检测细胞中周期蛋白D1、P16蛋白表达情况。结果DHA对人胃癌细胞具有时间、浓度依赖性抑制作用;且浓度依赖性阻滞细胞周期,使G0/G1期细胞增多,S期细胞减少(P<0.05)。DHA浓度依赖性下调cyclin D1蛋白,上调p16蛋白的表达(P<0.05)。结论DHA通过下调cyclin D1蛋白,上调p16蛋白的表达,调节细胞周期调控通路,阻滞细胞于G0/G1期,有效抑制人胃癌细胞的增殖。 Objective To investigate the inhibitory effect of dihydroartemisinin(DHA)on the proliferation of human gastric cancer cell line BGC-823 and its effect on the protein expression of cyclin D1 and P16.Methods The human gastric cancer cell line BGC-823 was intervened by 25-100μmol/L DHA,and the cell proliferation rate was determined by MTT colorimetry.AO-EB staining was used to determine the cell apoptosis,flow cytometry was used to determine the cell cycle,and Western blot ting was used to determine the protein expression of cyclin D1 and P16.Results DHA inhibited human gastric cancer cells in a time-and concentration-dependent manner,and blocked cell cycle in a concentration-dependent manner,resulting in an increase in G0/G1 phase cells and a decrease in S phase cells(P<0.05).DHA down-regulated Cyclin D1 protein expression,and up-regulated P16 protein expression in a concentration-dependent manner(P<0.05).Conclusion DHA may effectively inhibit the proliferation of human gastric cancer cells by down-regulating Cyclin D1 protein expression,up-regulating P16 protein expression,regulating cell cycle for pathway regulation and blocking cells at G0/G1 phase.
