微量滴定蒽酮法测定甜玉米芯可溶性糖含量方法的建立 被引量:19

Determination of soluble sugar from sweet corn cob by microtiter method of the anthrone-sulfuric acid colorimetric assay

摘要 以甜玉米芯为原料,利用单因素及响应面实验,优化微量滴定蒽酮-硫酸法测定甜玉米芯可溶性糖含量,所优化方法与苯酚-硫酸法测定结果无差异,操作简便快速,具有良好的线性、稳定性、重复性及精密度。结果表明,微量滴定蒽酮法最佳测定条件为:料液量50μL、硫酸浓度90%、蒽酮用量0.2 g、加热温度91℃、加热时间15.4 min,在此条件下测得甜玉米芯中可溶性糖含量为20.8%。该方法快速安全、节约试剂,适用于高通量样品测定。 In this experiment, we optimized the conditions for the determination of soluble sugar in sweet corn cob by microtitration of anthrone. The optimized method was compared with the traditional phenolsulfuric acid method, and the results showed no difference. The method is simple and fast, with good linearity, stability, repeatability and precision. Response surface methodology was used to optimize the optimal conditions for the determination of soluble sugar in sweet corncob. The optimum conditions were as follows: material liquid volume 50 μL, anthrone dosage 0.2 g, sulfuric acid concentration 90%, heating temperature 91 ℃, heating time 15.4 min. Under these conditions, the soluble sugar content in sweet corncob was 20.8%. Compared with the traditional method, the micro-titration method is simple, fast, safe and reagent-saving. It is suitable for the determination of high-throughput samples.
