苹果链格孢菌胞外多糖及分光光度法改用酶标仪法的稳定性 被引量:9

Stability of the Alternaria mali Roberts Exopolysaccharides and the Change from Spectrophotometer to Multiskan Spectrum

摘要 通过对传统多糖含量测定方法蒽酮法加以改进,以全波长酶标仪代替可见分光光度计进行糖含量测定,对苹果链格孢菌胞外多糖在不同温度、光照、pH值和金属离子条件下的此测定方法稳定性进行研究。结果表明,分光光度法改用酶标仪法后,其线性关系良好、稳定性及精密度较好,具有可重复性,且操作简便,适用于大样本测量分析。苹果链格孢菌胞外多糖在20~80℃下的热稳定性较好,在pH=6的弱酸及Na^+溶液中多糖稳定性较好,强酸强碱及Cu^(2+)、Fe^(3+)溶液中稳定性较差,光照可对多糖稳定性产生影响。 In order to find an efficient, quick and stable method for determination of A lternaria mali Roberts exopolysaccharides, the content and stability of exopolysaccharides in light condition,different temperature,pH,metal ions were studied by a muhiskan spectrum microplate spectrophotometer. The results showed that the method for mensurating the exopolysaccharide content by muhiskan spectrum had good linear relationship, stability and repeatability,the operation was simple and suitable for large sample measurement and analysis. The Alternaria mdi Roberts exopolysaccharides had better thermal stability at 20-80℃ and remained stable in weak acid (pH=6) and Na^+ ion solution,strong acid and alkali solution or Cu^2+,Fe^3+ ions had an impact on the stability of exopolysaeeharides,the light could also influence the stability.
