摘要
【目的】建立液相色谱-质谱联用测定果葡糖浆中葡萄糖、果糖含量的方法.【方法】采用BEH Amide色谱柱(2.1 mm×100 mm,1.7μm)分离,以20 mmol/L甲酸铵水溶液∶乙腈为流动相(25∶75,V/V),采用加热电喷雾离子源,选择负离子采集方式进行检测,样品经过滤后直接上机测试.【结果】该方法测定葡萄糖的线性范围为1.56~31.2μg/mL,线性相关系数R 2=0.9998,检出限为0.32μg/mL;果糖的线性范围为6.5~130μg/mL,线性相关系数R 2=0.9988,检出限为0.74μg/mL.【结论】本方法操作简便、分析速度快、灵敏度高,可用于果葡糖浆中糖类物质含量的测定.
【Objective】A liquid chromatography-mass spectrometry(LC-MS)method was developed for the determination of glucose and fructose in fructose syrup.【Method】The chromatographic separation was achieved on a BEH Amide chromatographic column(2.1 mm×100 mm,1.7μm),with a mobile phase composed of 20 mmol/L ammonium formate aqueous solution and acetonitrile(25∶75,V/V).The mass spectrometer was operated in the negative ion mode using heated electrospray ion source.The samples were detected directly after filtering.【Result】The linear range of glucose was 1.56~31.2μg/mL,the linear correlation coefficient was 0.9998 and the detection limit was 0.32μg/mL.The linear range of fructose was 6.5~130μg/mL,the linear correlation coefficient was 0.9988,and the detection limit was 0.74μg/mL.【Conclusion】This method was simple,rapid,and sensitive,and could be used for the determination of sugar contents in fructose syrup.
