Study on cultivation technology of high quality Cordyceps militaris

摘要 由于冬虫夏草野生资源匮乏,且现在还无法进行人工培育,而蛹虫草由于其药理作用与冬虫夏草相似,并且已经实现了大规模生产,因此蛹虫草便成为了冬虫夏草的替代品。以实验室保存的蛹虫草菌种为研究对象,采取单因素试验对蛹虫草固体培养基进行优化,通过对其在固体培养基的生长情况、对子实体多糖含量的测定、抗氧化活性的评价最终得到蛹虫草固体培养的最优碳源、氮源以及无机盐。经过单因素试验筛选得到蛹虫草高品质子实体栽培营养条件。蛹虫草固体子实体生长的最佳碳源、氮源、无机盐分别为蔗糖、酵母浸膏、磷酸二氢钾、氯化钙;利于子实体中多糖积累的碳源依次为:麦芽糖>蔗糖>葡萄糖;氮源为:牛肉浸膏>酵母浸膏>蛋白胨;无机盐依次为:氯化钙>氯化镁>磷酸氢二钾。从抗氧化活性评价结果可得:酵母浸膏>蛋白胨;氯化钠>硫酸镁>氯化钙>氯化镁。采用单因素实验确立了蛹虫草CMKN菌株的固体栽培配方,为蛹虫草的高品质子实体栽培生产提供理论依据及前期技术基础。 Due to the lack of wild resources in Cordyceps,it is still unable to carry out artificial cultivation,and Cordyceps sinensis is similar to Cordyceps,and has achieved large-scale production,and therefore,the cordona will become a replacement of Cordyceps.Methods:based on the cordyceps militaris strains preserved in our laboratory as the research object,adopt single factor experiment optimize the cordyceps militaris solid medium,by its growth in the solid medium,determination of the content of polysaccharide subsidiary entities,antioxidant activity evaluation of cordyceps militaris solid cultivation,the optimal carbon source,nitrogen source and inorganic salts.Results:The nutritional conditions of high quality fruiting body of Cordyceps militaris were obtained by single factor test.The best carbon source,nitrogen source and inorganic salt for solid fruiting body growth of Cordyceps militaris were sucrose,yeast extract,potassium dihydrogen phosphate and calcium chloride.The carbon sources conducive to the accumulation of polysaccharides in the fruiting body were:maltose>sucrose>glucose;The nitrogen source was:beef extract>yeast extract>peptone;Inorganic salts in order:calcium chloride>magnesium chloride>dipotassium hydrogen phosphate.From the results of antioxidant activity evaluation,yeast extract>peptone was obtained.Sodium chloride>magnesium sulfate>calcium chloride>magnesium chloride.Conclusions:In this experiment,single factor experiment was used to establish the solid cultivation formula of CMKN strain of Cordyceps militaris,which provided theoretical basis and early technical basis for high quality fruitbody cultivation and production of Cordyceps militaris.
